摘要 目的探讨JAK/STAT信号通路在大鼠变应性鼻炎(AR)鼻黏膜中的表达,探讨其在变应性鼻炎中的作用。方法 40只大鼠分为两组:对照组和变应性鼻炎组。变应性鼻炎组采用二异氰酸甲苯酯(TDI)制作大鼠实验性AR模型;对照组则给予TDI溶剂醋酸乙酯滴鼻。取两组大鼠下鼻甲组织,利用实时荧光定量PCR(q-PCR)检测JAK1、JAK2、STAT1和STAT3 mRNA表达,利用Western blot检测p-JAK1、p-JAK2、p-STAT1和p-STAT3蛋白表达。结果q-PCR结果显示,变应性鼻炎组JAK1、JAK2、STAT1和STAT3 mRNA表达均显著高于对照组(P<0.05);Western blot结果显示,变应性鼻炎组p-JAK1、p-JAK2、p-STAT1和p-STAT3蛋白表达均显著高于对照组(P<0.05)。结论变应性鼻炎发生后JAK/STAT信号通路被激活,可能是变应性鼻炎治疗的靶点。 Objective To explore the expression of JAK/STAT signaling pathway in nasal mucosa of allergic rhinitis rats and its underlying mechanism. Methods Model of experimental allergic rhinitis was made by toluene diisocyanate, and 40 rats were divided into 2 groups: the control group and the allergic rhinitis group. The mRNA expression of JAK1, JAK2, STAT1 and STAT3 were detected by real time PCR. Meanwhile, the protein expression of p-JAK1, p-JAK2, p-STAT1 and p-STAT3 were valued by western blot. Results Real time PCR showed that the mRNA expression of JAK1, JAK2, STAT1 and STAT3 in the allergic rhinitis group were significantly higher than that in the control group ( P 〈0.05). Meanwhile, western blot showed that, the protein expression of p-JAK1, p-JAK2, p-STAT1 and p-STAT3 were markedly higher than that in the control group ( P 〈0.05). Conclusion JAK/STAT signaling pathway might be activated in allergic rhinitis rats, which could be the target of the treatment of allergic rhinitis.
