阴虚水肿证大鼠模型的建立及猪苓汤的干预作用 被引量:6

Establishment of Rat Model of Yin Deficiency and Edema Syndrome and Intervention Effect of Zhuling Decoction

摘要 目的:评价并探讨一种类似阴虚水肿证大鼠模型制作方法并观察猪苓汤的干预作用。方法:100只雄性SPF级Wistar大鼠分为空白组(不做处理)、对照组(尾静脉注射生理盐水)、阿霉素组(尾静脉注射阿霉素),2周后24小时尿蛋白定量≥100 mg提示阿霉素肾病模型复制成功,弃去对照组、造模失败及死亡大鼠,重新分为空白组、阿霉素模型组、阴虚水肿组(阿霉素模型基础上连续灌服左甲状腺素钠片悬液)、猪苓汤组(阴虚水肿组基础上连续灌服猪苓汤)。观察各组大鼠不同时间点皮肤含水量、肛温、甲状腺功能等变化,以猪苓汤干预反证该模型是否成立。结果:与空白组比较,阿霉素组大鼠第2周阴囊水肿明显,第4周皮肤含水量高于空白组(P<0.05);连续灌胃左甲状腺素钠片2周阴虚水肿组大鼠活动频繁,躁动不安,易激惹,饮水量多,鼠笼垫料潮湿,排泄物增多,脱毛,四肢及阴囊水肿明显,肛温、总三碘甲状腺原氨酸(total triiodothyronine,TT3)、血清总甲状腺素(total thyroxine,TT4)高于空白组、阿霉素组(P<0.05);连续猪苓汤灌胃4周,猪苓汤组大鼠肛温低于阴虚水肿组(P<0.05);连续猪苓汤灌胃8周,猪苓汤组尿量多于阿霉素组、阴虚水肿组(P<0.05),连续猪苓汤灌胃8周,猪苓汤组与阴虚水肿组比较,TT3、TT4、TSH差异无统计学意义(P>0.05)。结论:一次性尾静脉注射阿霉素结合连续左甲状腺素钠片灌胃可一定程度模拟阴虚水肿状态;猪苓汤可改善阴虚水肿证大鼠躁动不安,降低阴虚水肿证大鼠肛温,增加尿量。 Objective :To evaluate and explore a method in establishing a rat model of Yin deficiency and edema Syndrome and to observe the intervention effect of Zhuling Decoction. Methods : 100 male SPF grade Wistar rats were randomly divided into blank group (no treatment), control group (injecting saline into the tail vein) and adriamycin group (injecting adriamycin into the tail vein). Two weeks later,the 24 h urine protein quantitation was more than or equal to 100 mg indicating that the adriamycin ne- phropathy model was successfully established, and the control group, model failure ones and death rats were abandoned. The rest rats were randomly divided into blank group, adriamycin model group and Yin deficiency and edema group (adriamycin model on the basis of continuous perfusion of Euthyrox) and Zhuling Decoction group (continuous perfusion of Zhuling Decoction was used to the Yin deficiency and edema group). The changes of skin water content, anal temperature and thyroid function were observed at different time points in each group. The establishment of the model was proved by the intervention of Zhuling Decoction. Results : Compared with the blank group, Scrotal edema in the adriamycin group was obvious in second weeks, and the skin water content in fourth weeks was significantly higher than that in the blank group (P 〈 0.05 ). Continuously administion Euthyrox for 2 weeks to the Yin deficiency and edema group, and the rats have shown obvious activities, restlessness, irritability, drinking water, squirrel cage material moist,increased excretion, hair removal, obvious extremities and scrotal edema, and the anal temperature Total triio- dothyronine (TT3) and total thyroxine (TT4) were significantly higher than that of the blank group and doxorubicin group (P 〈 O. 05 ). Zhuling Decoction was administered continuously for 4 weeks, and the anus temperature of the rats in the phlegm group was lower than that in the group of Yin deficiency and edema (P 〈 0.05 ). The continuous Zhuling Decoction was administered for 8 weeks. The amount of urine in the Zhuling Decoction group was significantly higher than that in the adriamycin group and the Yin deficiency and edema group (P 〈 0.05 ), Zhuling Decoction was administered for continuous 8 weeks and there was no significant difference in TT3, TT4 and TSH between Zhuling Decoction group and Yin deficiency and edema group (P 〉 0.05 ).
