摘要
目的 探讨CR3-TLR2合作体在小鼠巨噬细胞识别马尔尼菲蓝状菌中的作用.方法 以小鼠巨噬细胞系RAW264.7为靶细胞,siRNA靶向下调巨噬细胞CR3、TLR2的表达,与马尔尼菲蓝状菌分生孢子共培养,流式细胞技术检测共培养前后巨噬细胞的吞噬率,ELISA检测共培养上清细胞因子水平.结果 siRNA靶向下调RAW264.7细胞CR3、TLR2、CR3+TLR2后,与马尔尼菲蓝状菌共培养,巨噬细胞对马尔尼菲蓝状菌分生孢子的吞噬率均下降(P<0.05),且以干扰CR3+TLR2后下降最明显(P<0.05);同时,巨噬细胞分泌细胞因子的水平下降(P<0.05).结论 在先天性免疫早期阶段,CR3-TLR2合作体为巨噬细胞识别、介导吞噬马尔尼菲蓝状菌的主要模式识别受体之一;IL-2、IFN-γ、IL-4、IL-10等Th1型和Th2型细胞因子均参与巨噬细胞抗马尔尼菲蓝状菌感染免疫.
Objective To evaluate the role of complement receptor 3-Toll like receptor 2 on murine macrophage in recognition of Talaromyces marneffei.Methods The siRNAs targeting interfere with CR3 and/ or TLR2 on mouse macrophage cell line RAW264.7,targeting down the expression of CR3,TLR2,CR3-TLR2,and then co-cultured with inactivated Talaromyces marneffei conidia.Phagocytosis rate was detected by flow cytometry,culture cytokine level was detected by ELISA.Results CR3,TLR2,CR3-TLR2 on RAW264.7 cells was interfered by siRNA,then co-cultured with inactivated Talaromyces marneffei conidia,all of the phagocytic rate decreased (P〈0.05),and the interfering with CR3+TLR2 was obvious (P〈0.05).Meanwhile,the cytokines secreted by macrophages decreased (P〈0.05).Conclusions In early stage of innate immunity,CR3-TLR2 collaboration on macrophage was one of the major pattern recognition receptors that participated recognition and mediated phagocytosis of Talaromyces marneffei.Thl-type cytokines and Th2-type cytokines of IL-2,IFN-γ,IL-4,IL-10 were involved in anti-infection immunity of macrophage resistance to Talaromyces marneffei.
