摘要
目的:采用同位素相对标记与绝对定量技术(TMT)联合二维液相色谱-串联质谱(2D-LC-MS/MS),筛选系统性红斑狼疮(Systemic Lupus Erythematosus,SLE)合并激素性股骨头坏死(steroid induced osteonecrosis of the femoral head,SONFH)筋脉瘀滞证、肝肾亏虚证患者与健康人之间的血清差异蛋白质,初步寻找出两种不同证型的潜在生物学标志物。方法:分别收集SONFH筋脉瘀滞证患者、肝肾亏虚证患者及健康人各5例血清,应用TMT联合2D-LC-MS/MS检测技术,寻找并鉴定差异表达的蛋白质。结果:共筛选出IGHV3-7、IGHV1OR15-1、ABCB9、FGB、F13A1、FGG、FGA SAA1、ACTB、CRP共10个表达差异倍数在两倍以上显著表达的蛋白质。其中IGHV3-7、IGHV1OR15-1、ABCB9、FGB、F13A1、FGG、FGA筋脉瘀滞组中显著表达;SAA1、ACTB、CRP肝肾亏虚组中显著表达。结论:IGHV3-7、IGHV1OR15-1、ABCB9、FGB、F13A1、FGG、FGA可能是SLE合并SONFH筋脉瘀滞证潜在的血清特异性蛋白标志物;SAA1、ACTB、CRP可能是SLE合并SONFH肝肾亏虚证潜在的血清特异性蛋白标志物。
Objective:To screen SLE patients with steroid-induced osteonecrosis of the femoral head(SONFH) stasis syndrome by using 2 D-LC-MS/MS with isotope relative labeling and absolute quantification technique TMT. Theserum differential proteins between patients with deficiency syndrome and healthy individuals were initially searched for potential biomarkers of two different syndromes. Methods:Five cases of patients with SONFH stasis syndrome, five cases of liver and kidney deficiency syndrome patients and five cases of healthy people were collected respectively.TMTand 2 D-LC-MS/MS were used to differentially identify expressed proteins.Result:A total of 10 proteins including IGHV3-7, IGHV1 OR15-1, ABCB9,FGB,F13 A1,FGG, FGA SAA1,ACTB and CRP were screened at more than two times. Among them,IGHV3-7, IGHV1 OR15-1, ABCB9, FGB, F13 A1,FGG and FGA were significantly expressed in the tendons with stamina and ventral stasis. SAA1,ACTB and CRP were significantly expressed in the liver-kidney deficiency group.Conclusion:IGHV3-7, IGHV1 OR15-1, CLEC3 B, FGB, F13 A1, FGG and FGA may be potential serum specific protein markers for SLE combined with SONFH tendon stasis syndrome. SAA1, ACTB and CRP may be SLE combined SONFH liver-kidney deficiency syndrome potential serum-specific protein markers.
