摘要 目的:探讨胶原酶消化法从人脂肪组织中提取脂肪干细胞(adipose-derived stem cells,ADSCs)的可行性。方法:采用胶原酶消化法将真空抽脂术抽取的新鲜脂肪组织进行消化分离,获取ADSCs并进行培养,用流式细胞术鉴定ADSCs的表面特异性标志物CD34、CD44、CD45、CD105的表达。结果:利用胶原酶消化法可成功提取ADSCs,其标志物CD44和CD105表达阳性,CD34为低表达, CD45表达为阴性。结论:通过胶原酶法可成功提取人ADSCs。 Objective: To investigate the isolation, culture and identification of the adipose-derived stem cells (ADSCs) from human adipose tissue in vitro by collagenase digestion method. Methods: The subcutaneous adipose tissue was obtained from caesarean section, and then was digested with 0.2% I collagenase under the aseptic condition. The isolated ADSCs were cultured and the morphology and proliferation charaeteristics were observed. ADSCs were identified by the expression of sin-face specific markers CD34, CD44, CD45 and CD105 with flow cytometry. Results: ADSCs were successfully isolated and cultured. The cell surface markers CD44 and CD105 were positively expressed, CD34 was weakly expressed, and CD45 were negatively expressed. Conclusion : The human ADSCs can be successfully isolated by collagenase digestion method.
