摘要
目的 探讨姜黄素对急性髓性白血病细胞 (HL- 6 0 )增殖和细胞周期的调控及细胞凋亡的影响。方法 采用 MTT法测定姜黄素在不同浓度、不同时间对 HL- 6 0的抑制作用 ;流式细胞仪分析细胞周期分布 ;透射电镜观察细胞的超微结构变化。结果 姜黄素可抑制 HL- 6 0细胞的生长 ,其抑瘤率与药物浓度和作用时间呈依赖关系 ;中效浓度 (IC50 )为 2 4 .8μm ol/ L 时 ,流式细胞仪分析证实姜黄素能使 HL- 6 0细胞聚积在 S期、G2 / M期 ;电镜观察发现姜黄素可使 HL- 6 0细胞超微结构发生改变。结论 姜黄素可干扰 HL- 6 0细胞的周期分布 。
Objective To investigate the impact of curcumin on the proliferation,cell cycle distribution, and apoptosis of acute myeloid leukemic cells HL 60. Methods The MTT method was used to test the inhibitive effects of curcumin on HL 60 in a range of doses and exposure periods. The Flow cytometic analysis was used to detect the cell cycle distribution. The cell ultrastructure was observed under the electronic microscopy. Results The curcumin inhibited the HL 60 cell effectively in a dose and time dependent manner, with an IC 50 of 24 8 μmol/L. The growth of HL 60 cell was arrested at the S and G 2/M stages. The change of the ultrastructure of HL 60 cells was detected. Conclusion Curcumin could interrupt the cell cycle and play a role in the antiproliferation of HL 60 cells and inducing apoptosis.
