摘要
目的 观察地西泮对芬太尼镇痛效应的影响 ,并初步探讨其可能的作用机制。方法 雌性昆明种小鼠6 0只 ,随机分为 6组 ,每组 10只 ,以热板法测定各组动物腹腔注射 (ip .)用药前后的痛阈。结果 2mg kg地西泮对小鼠的基础痛阈无明显影响 ,4 0mg kg芬太尼可明显延长小鼠的痛阈 (P <0 .0 1) ,2mg kg地西泮可显著拮抗 4 0mg kg芬太尼的镇痛效应 (P <0 .0 1) ,而苯二氮受体阻断剂———氟马泽尼 5mg kg可抵消地西泮对芬太尼镇痛效应的拮抗作用 ,单用氟马泽尼可明显增强芬太尼的镇痛作用 (P <0 .0 1)。结论 地西泮可拮抗芬太尼的镇痛效应 ,其机制可能与其激动苯二氮受体后增强γ -氨基丁酸的作用有关。
Objective To study the influence of diazepam on the analgesic effect of fentanyl in mice.?Methods Hot plate test was used to determine the pain threshold before and after the application of drugs ip.?Results Diazepam (2 mg/kg) showed no effect on the basic pain threshold of the mice. Fentanyl (40 mg/kg) could increase the pain threshold ( P <0.01) and diazepam antagonized the analgesic effect of fentanyl significantly ( P <0.01). Flumazenil (5 mg/kg), the blocker of benzodiazepine receptor, could conceal the antagonistic effect of diazepam and potentiate the analgesic effect of fentanyl ( P < 0.01). ?Conclusions Diazepam can antagonize the analgesic effect of fentanyl, possibly by activating benzodiazepine receptors and enhancing the inhibitory action of GABA.
