摘要
目的研究红花注射液(SY)对控制性降压(CH)诱导的缺血缺氧性损伤兔海马CA1神经元nNOS表达的影响。方法构建缺血缺氧性脑损伤动物模型,SY组在降压前30min静脉注射红花注射液2mL/kg(1mL含SY1.6mg)。nNOS免疫组化染色观察CA1神经元形态学改变,Westernblotting检测nNOS蛋白的表达水平。结果SY组nNOS免疫组化染色的光密度值显著低于CH组(P<0.05),其nNOS蛋白表达水平也明显低于CH组,但高于Normal组(P<0.05)。结论红花注射液显著减轻CH诱导的兔海马缺血缺氧性损伤,可能是通过抑制nNOS蛋白表达起作用的。
Objective To study the effect of safflower injection ( SY) on nNOS expression of hippocampal CA1 neuronssuffering from hypoxic - ischemic injury induced by controlled hypotension ( CH) in rabbits. Methods The hypoxic -ischemic brain injury model was constructed and the SY group was intravenously injected with 2 mL/kg of safflower injection(1 mL, containing 1.6 mg SY) 30 min before CH. The neuronal changes in hippocampal CA1 were observed for nNOS immunohistochemicalstaining, and western blotting was used to detect the expression of nNOS. Results Optical density valuesof nNOS staining in the SY group were significantly lower than that in the CH group (P <0. 05) . The expression of nNOS inthe SY group was significantly lower than that in the CH group, but it was higher than that in the normal control group (P <0. 05). Conclusion SY can significantly attenuate the hypoxic - ischemic hippocampal injury, possibly by inhibiting nNOS expression.
