尖锐湿疣患者外周血Treg/Th17细胞平衡及相关转录因子的表达

Balance of Treg/Th17 cells and the expression of related transcription factors in peripheral blood of patients with condyloma acuminatum

目的探讨Treg/Th17细胞失衡在尖锐湿疣(CA)发病及复发机制中的作用及意义。方法选取52例CA患者(初发27例,复发25例)作为研究组,30例健康者为正常对照组,采用流式细胞术(FCM)检测其外周血Treg、Th17细胞的比例,实时荧光定量PCR检测外周血单个核细胞中Foxp3 mRNA和RORγt mRNA的表达水平。结果 CA患者外周血Treg细胞比例、Foxp3 mRNA表达水平高于正常对照组,复发组高于初发组,差异均有统计学意义(均P<0.05);Th17细胞比例、RORγt mRNA表达水平低于正常对照组,复发组Th17细胞比例低于初发组,差异有统计学意义(均P<0.05);Treg/Th17比值高于正常对照组[4.60(3.20,8.68)vs 1.39(1.05,2.05)],复发组高于初发组[8.19(4.21,10.81)vs 3.52(2.47,4.85)],差异均有统计学意义(均P<0.05)。结论 CA患者存在Treg/Th17细胞失衡,复发患者Treg/Th17细胞失衡更显著,并可能在CA发病及复发机制中起重要作用。

ObjectiveTo evaluate the role and significance of Treg/Th17cells imbalance in pathogenesis and recurrence mechanism of condyloma acuminatum(CA).Methods52patients with CA were selected as study group(CA group,27with initial occurrence of CA,25with recurrence of CA),30healthy persons were as control group,flow cytometry was used to detect the proportion of Treg cells and Th17cells in the peripheral blood,the expression level of Foxp3mRNA and RORγt mRNA in peripheral blood mononuclear cells was determined by real time quantitative polymerase chain reaction.ResultsThe proportion of Treg cells and the expression level of Foxp3mRNA in peripheral blood in CA group was higher than that in control group,recurrence CA group was higher than initial occurrence CA group,difference was significant(both P<0.05);the proportion of Th17cells and expression level of RORγt mRNA in CA group was significantly lower than that in control group,proportion of Th17cells in recurrence CA group was lower than initial occurrence CA group,there was significant difference(both P<0.05).The proporation of Treg/Th17in CA group was higher than that in healthy controls(4.60[3.20,8.68]vs1.39[1.05,2.05],P<0.05),recurrence CA group was higher than initial occurrence CA group(8.19[4.21,10.81]vs3.52[2.47,4.85],P<0.05).ConclusionThere is an imbalance between Treg cells and Th17cells in patients with CA,especially in patients with reccurrence of CA,the imbalance of Treg/Th17cells may play an important role in the pathogenesis and recurrence mechanism of CA.
