摘要
目的检测阿尔茨海病(Alzheimer’s disease,AD)患者血清长链非编码RNA(LncRNA)BC200表达水平,分析其与患者临床特征的关系,探讨其在AD诊断中与血管性痴呆(vascular dementia,VaD)鉴别诊断中的价值。方法采用实时荧光定量聚合酶链反应对41例AD患者、37例VaD患者、40例对照组患者血清中的LncRNA BC200表达水平进行检测。同时采用ELISA法检测各组患者脑脊液中Aβ1-42蛋白和Tau蛋白水平,分析LncRNA BC200、Aβ1-42和Tau在3组间进行鉴别诊断的价值。应用ROC曲线分析LncRNA BC200、Aβ1-42和Tau诊断AD的敏感度和特异度。结果LncRNA BC200表达水平和AD病程相关(P<0.05),而与性别、年龄和受教育程度均无显著相关(均P>0.05)。AD组患者的LncRNA BC200和Tau水平均明显高于VaD组和对照组,Aβ1-42水平显著低于VaD组和对照组(均P<0.01);VaD组患者的LncRNA BC200和Aβ1-42水平与对照组比较,差异无统计学意义(均P>0.05),而Tau水平显著高于对照组(P<0.01)。LncRNA BC200、Aβ1-42和Tau三者联合检测的敏感度(88.5%)、特异度(91.3%)和曲线下面积AUCROC(0.965)均比单独检测高,具有较好的诊断价值。结论 AD患者血清中LncRNA BC200表达水平明显升高,LncRNA BC200可用于AD和VaD的鉴别诊断,LncRNA BC200有望成为AD诊断的潜在生物标志物。
Objective To detect the scrum levels of longnoncoding RNA(LncRNA.)BC200 inpatients with Alzheimer’s discasc(AD),analyze its relationship with clinical features,and explore its diagnostic value for AD and its differential diagnostic value between AD and vascular dementiaCVaD).Methods Scrum levels of LncRNA BC200 were detected by real-time quantitative polymerase chain reaction in 4 1 AD patients,37 VaD patients,and 40 control patients.Meanwhile.the levels ofβ--amyloid peptidd42protcin(Aβ142)and tau protein(Tau)in cerebrospinal fluid were measured by ELISA.The diagnostic value of LncRNA BC200,A[i--2 and Tau in the differential diagnosis between AD and VaD was analyzed.T'he sensitivity and specificity of LncRNA BC200,AJ3-2 andTauwere analyzed by using the receiver operating charactcristic(ROC)curve.Results The expression level of LncRNA BC200 was correlated with the duration ofAD(P〈0.05.),but notwith gcndcr,age and educational level(P〉0.05).The levels of LncRNA BC200 andTauwere significantly higher inADgroup than inVaD group and control group,and the level ofA[-'i2 was significantly lower in ADgroup than in VaDgroup and control group(P〈0.05).The levels of Ln-eRNA BC200 and A[-i2 in VaDgroupwere not significantly different from those in control group(P〉0.05.),while the level of Tau was significantly higher inVaD group than in control group(P〈0.01).The sensitivity,specificity andAUCROCof the com bined detection of LncRNA BC200,Aβ1-42 andTauwere 88.5%,9.3%and 0 965,which were significantly higher than those of their single detection and showed a good diagnostic valie.Conclusion Scrum LncRNA BC200 levels were significantly increased in AD patients?which may be used as a differential diagnosis marker between AD and VaD.LncRNA BC200 is expected to be a potential biomarkcr forADdiagnosis.
