四神丸对慢性结肠炎大鼠结肠组织相关因子表达的影响

Expressions of SOD,MPO,MDA,T-AOC,NO and i-NOS from Colonic UC Rats Treated by Sishen Pills

目的:探究四神丸对慢性复发型结肠炎大鼠结肠组织超氧化物歧化酶(SOD)、髓过氧化酶(MPO)、丙二醛(MDA)、总抗氧化能力(T-AOC)、一氧化氮(NO)、一氧化氮合酶(i-NOS)表达的影响。方法:参考文献复制慢性复发型SD大鼠结肠炎模型,将60只大鼠随机平均分为正常组、模型组、四神丸高、中、低剂量组和美沙拉嗪对照组。观察大鼠一般情况,进行结肠肉眼下及镜下损伤评分,分别采用考马斯亮蓝法检测总蛋白量、化学比色法检测SOD、MPO、MDA、T-AOC、NO和i-NOS的表达量。结果:与模型组对比,四神丸各组大鼠结肠重量明显减轻,结肠重量指数明显下降,结肠长度增长,同时结肠肉眼下及镜下损伤评分均明显降低,而SOD和T-AOC的活性均显著升高,但MPO、MDA、NO和i-NOS的活性却显著降低。结论:四神丸可能通过恢复结肠黏膜氧化代谢平衡而达到改善或减轻慢性复发型UC大鼠的目的,但是否通过i-NOS/NO途径仍需进一步研究。

Objective:To observe the effect of Sishen Pills(SSP)on expressions of lsuperoxide dismutase(SOD),myeloperoxidase(MPO),malondialdehyde(MOA),total antioxidant capacity(T-AOC),nitric oxide synthase(NO)and nitric oxide synthase(i-NOS)in colonic tissues of rats with chronic recurrent colitis.Methods:Sixty rats were randomly divided into normal group,model group,high,medium and low doses of SSP groups and mesalazine positive control group.Macroscopic and microcosmic injury scores were analyzed,and the indexes of colonic weight,colonic weight and colonic length wre measured to evaluate the therapeutic effect of SSP treating chronic relapsing colitis.While the products of SOD,MPO,MDA,T-AOC,NO and i-NOS in colonic tissues were tested by chemical colorimetry.Results:Compared with model group,the colonic weight index,the macroscopic and microcosmic injury scores were obviously decreased in each groups of SSP,however,the colonic length was recovered.Meanwhile,after rats with chronic relapsing colitis treated by SSP,the concentrations of SOD and T-AOC were significantly increased,and the levels of MPO,MDA,NO and i-NOS were inhibited.Conclusion:Sishen Pills effectively alleviated the chronic relapsing UC by recovering the balance of oxidative metabolism in colonic mucosal tissues.However,further study is required if the i-NOS/NO pathway is adopted.
