两种高脂高糖饲料致SD大鼠Ⅱ型糖尿病模型的效果比较 被引量:2

Comparison of the Effects in SD Rat Experimental Type Ⅱ Diabetes Mellitus Model Induced by Two High-fat and High-glucose Diets

摘要 探讨不同高脂高糖饲料对SD大鼠Ⅱ型糖尿病动物模型的影响,分析模型鼠的主要死亡因素,以期提高造模成功率。选取用于复制SD大鼠Ⅱ型糖尿病动物模型的两种配方饲料,饲喂SD大鼠42 d后,以30 mg/kg腹腔注射链脲佐菌素(STZ)溶液,且连续观察14 d,分别在试验开始当天、第7天、第14天、第21天、第28天、第35天、第42天、第49天、第56天禁食12 h后,观察大鼠是否存在"多食、多饮、多尿、体重减轻"症状,测定大鼠体重、空腹血糖(FBG)等;在第42天、第56天禁食12 h后心脏采血,测定口服葡萄糖耐量(OGTT)及甘油三脂(TG)、胆固醇(TC)、低密度脂蛋白(LDL),高密度脂蛋白(HDL),空腹血浆胰岛素(FSI)的含量,并计算胰岛素敏感指数(ISI)及胰岛素抵抗指数(IRI)。两种高脂高糖配方饲料组在第42天、第56天,大鼠体重、OGTT、ISI均低于正常组,FBG、TG、FSI及IRI的值明显高于对照组,H1组、H2组成模率分别为75%和50%,死亡率分别为25%和50%。两种高脂高糖配方饲料均可复制大鼠Ⅱ型糖尿病模型,但H1组优于H2组。 To explore the effects of high-fat and high-glucose diets on the type Ⅱ diabetes mellitus in SD rat models and analyze the main death factors in the hope of improving the success rate of model.After the SD rat in the experimental groups were fed respectively with high-fat and high-sugar diets with varying amounts of lard and sugar,with 30 mg/kg intraperitoneal injection of streptozotocin (STZ) simultaneously at the 42th day, and they were continuously observed for 14 days. At the beginning of the experiment, on the 7th, 14th, 21st, 28th, 35th, 42nd, 49th and 56th days after fasting for 12 hours, we observed whether the rats had symptoms of “overeating,drinking,urination and weight loss” by measuring their body weight and fasting blood glucose (FBG) in all the groups.Oral glucose tolerance (OGTT), triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL), high-density lipoprotein cholesterol(HDL), fasting serum insulin (FSI) were measured and the insulin sensitive index (ISI) and insulin resistance index (IRI) were calculated on the 42th and 56th day. Weight,OGTT,ISI of the experimental groups rat were significantly lower than those in SD rats of the control group.FBG,TG,FSI and IRI of the experimental groups rat were significantly higher than those in SD rats of the control group and the success rate of the experimental H1 group and H2 group was 75% and 50%,respectively,and the death rate was 25% and 50%. Two high-fat and high-glucose formula feed could both duplicate type Ⅱ diabetes mellitus in SD rat models,but H1 group was superior to H2 group.
