PGE_2及其受体在小鼠月经模型中子宫内膜崩解期的含量与表达研究 被引量:1

Expression of PGE_2 and its receptor PTGER2 during endometrial breakdown in mouse menstrual-like model

摘要 目的探讨前列腺素E2(PGE2)及其前列腺素E2受体(PTGER2)在小鼠月经模型中的含量和表达变化。方法选取40只小鼠,建立小鼠月经模型,用ELISA方法检测孕酮撤退后0~24h小鼠子宫组织匀浆中PGE2的含量;用实时定量PCR、免疫组化和Western blot方法检测子宫内膜崩解期Ptger2mRNA和蛋白质在子宫组织中的表达与定位。结果 PGE2的含量在孕酮撤退崩解期子宫组织中呈现逐渐升高的趋势,16h达到最大(P<0.01);PTGER2蛋白质在孕酮撤退的0~24h定位于整个蜕膜化区域且未呈现区域性的变化;Ptger2mRNA在孕酮撤退后0~16h,无显著变化(P>0.05),在孕酮撤退后24h其表达较0~16h显著升高(P<0.01),但PTGER2蛋白质在孕酮撤退后0~24h的表达无显著变化(P>0.05)。结论提示PGE2对子宫内膜的崩解剥脱作用可能通过PGE2含量的增加,而不是通过其受体PTGER2表达的增加来实现。 Objective: To explore the expression of PGE2 and its receptor PTGER2 during endometrial breakdown in mouse menstrual-like model.Methods:Forty mice were selected to establish the mouse menstrual-like model.The content of PGE2 during endometrial breakdown by ELISA was assayed.The expression and localization of Ptger2 mRNA and protein in uterine tissue during endometrial disintegration were detected by real-time quantitative PCR,immunohistochemistry and Western blot.Results:The content of PGE2 in the uterus was increased during the endometrial breakdown,and reached the maximum at 16 hours after progesterone withdrawal(P<0.01).PTGER2 protein was located in the whole decidual zone and did not show the regional zone change during the endometrial breakdown(P>0.05).Ptger2 mRNA showed no changes from 0-16 hours and significantly increased at 24 hours after progesterone withdrawal(P <0.01).However,PTGER2 protein showed no significant change during endometrial breakdown(P>0.05).Conclusions:The breakdown and shedding of endometrium may be mediated by an increase in the PGE2 content,rather than by an increase in the expression of its receptor PTGER2.
