摘要
目的检测LINC00152在胰腺癌多个细胞系及组织中的表达,并探讨其表达水平与胰腺癌患者临床、病理特征之间的关系。方法实时荧光定量PCR(qRT-PCR)检测LINC00152在6株人胰腺癌细胞系和1株人胰腺导管上皮细胞中的表达,同时检测在100例胰腺癌组织及相其配对的相邻的非癌组织中LINC00152的表达情况,并分析LINC00152的表达水平与胰腺癌患者临床病理特征及生存预后之间的关系。结果 qRT-PCR显示,相对于人胰腺正常细胞系(HPDE),LINC00152的表达水平在6株人胰腺癌细胞系(PANC-1,MIA PaCa-2,Panc03.27,AsPC-1,SW1990,CFPAC-1)中明显地升高(均P <0.01);在人胰腺癌临床组织样本中,癌组织的LINC00152的表达水平明显高于癌旁组织(P <0.01),胰腺癌中LINC00152表达水平与淋巴结转移及临床分期密切相关(均P <0.05),而与患者的性别、年龄以及患者肿瘤的直径大小无关(均P> 0.05),Kaplan-Meier生存分析结果显示,相对于LINC00152低表达的患者,高表达患者的预后差(P <0.05)。结论 LINC00152的表达量在胰腺癌组织和细胞中异常增高,与胰腺癌的发生发展相关,可能是一个胰腺癌诊断以及治疗的有效靶点。
Objective To detect the expression of LINC00152 in pancreatic cancer tissues and cells,and to analyze its relationship with clinicopathological features and prognosis. Methods Real-time PCR(qRT-PCR) was used to detect LINC00152 expression in 100 cases of pancreatic carcinoma tissues and matched paracancerous tissues,6 pancreatic cancer cell lines and 1 normal pancreatic epithelial cell lines;the relationship between the expression level of LINC00152 in pancreatic cancer and pancreatic cancer clinical pathological parameters was analyzed. Results The results of q RT-PCR showed that the expression level of LINC00152 in the human pancreatic cancer cell lines PANC-1,M IA PaCa-2,Panc 03.27,AsPC-1,SW1990 and CFPAC-1 was higher than that in the human esophageal normal cell line HPDE(P < 0.01). The expression of LINC00152 in the pancreatic cancer cell carcinoma was higher than that in the adjacent tissues(P < 0.01). The expression of LINC00152 in the pancreatic cancer cell carcinoma was closely related to lymph node metastasis and clinical stage(all P < 0.05), but was not related to sex,age, tumor diameter(P > 0.05). Kaplan-Meier survival analysis showed that the prognosis of patients with high LINC00152 expression was poor(P < 0.05). Conclusion LINC00152 is related to the occurrence and development of pancreatic cancer. It may be a new target of gene diagnosis and therapy of pancreatic cancer.
