摘要 为研究扩展莫尼茨绦虫分离株核糖体ITS序列的遗传变异情况,并利用ITS序列探讨扩展莫尼茨绦虫与其它带科绦虫的种群发育关系,本研究利用PCR对扩展莫尼茨绦虫分离株核糖体ITS序列进行扩增、测序及分析。结果表明:16株扩展莫尼茨绦虫分离株核糖体ITS序列长度一致,为1462~1473bP,且分离株与基因库扩展莫尼茨绦虫位于同一大分支,可以与其它带科绦虫有效鉴别。说明ITS序列种内较为保守,种间差异较大,可以作为扩展莫尼茨绦虫的种间遗传变异研究的分子标记。 To analyze sequence variations in the ribosomal inter-transcribed spacer( ITS) among M.expansa isolates in Hunan Province,and explore phylogentic relationship between M. expansa and other tapeworm,the ITS regions of ribosomal DNA of M.expansa obtained were amplified by PCR,sequenced and analyzed. The results showed that the lengths of ITS sequences of 16 M.expansas isolates were 1 462-1 473 bp. The constructed phylogenetic tree based on ITS gene sequences revealed that the Hunan isolates and the M.expansa available in GenBank were clustered in the same clade. It is concluded that ITS sequences could be used as a genetic marker for interspecies genetic variation research of M.expansa.
