Simultaneous determination of three constituents in Qilin pills by HPLC

目的:建立HPLC法同时测定麒麟丸(制何首乌、墨旱莲、淫羊藿等)中3种成分的含量。方法:麒麟丸50%乙醇提取液的分析采用VensuilXBPC18(4.6mm×250mm,5μm);以含0.05%三氟乙酸的水溶液(A)和含0.05%三氟乙酸的乙腈溶液(B)为流动相,梯度洗脱;体积流量1.2ml/min;柱温30℃;检测波长220nm。结果:橙皮苷、丹酚酸B和淫羊藿苷分别在6~18μg/ml(r=0.9998),3~9μg/ml(r=0.9998),2~6μg/ml(r=0.9999)范围内线性关系良好,平均加样回收率分别为95.9%、93.6%、92.4%,RSD分别为1.6%、3.0%、4.6%。结论:该方法准确、重复性好,可用于质量控制。

Objective: To establish an HPLC method for the simultaneous determination of the contents of three constituents in Qilin pills (Polygoni Multiflori Radix Praeparata, Ecliptae Herba, Epimedii Folium). Methods: The analysis of 50% ethanol extract of Qilin pills was performed on a Vensuil XBP C18 (4.6 mm×250 mm,5 mm)by gradient elution with the mobile phase comprising of acetonitrile-0.05% trifluoroacetic acid at volume flow 1.2 ml/min, 30 ℃ and detection wavelength 220 nm. Results: Hesperidin, salvianolic acid B and icariin showed good linear relationship in the ranges of 6-18 mg/ml (r=0.999 8),3-9 mg/ml (r=0.999 8),2-6 mg/ml (r=0.999 9)with average recoveries of 95.9%, 93.6% and 92.4% and RSDs of 1.6%, 3.0% and 4.6%, respectively. Conclusion: This method is accurate and reproducible and can be used for the quality control of Qilin pills.
