EDTA对氯己定去除粪肠球菌生物膜效果的影响 被引量:2

摘要 目的 在体外环境下建立粪肠球菌(E.faecalis)生物膜,观察乙二胺四乙酸二钠(EDTA)作为分散剂对氯己定(CHX)清除生物膜效果的影响。方法 通过96孔板建立体外24h粪肠球菌生物膜模型,将生物膜随机分为A、B、C、D、E、F、G7组,A—E组分别加入1、2、4、8、16 MIC(最低抑菌浓度)EDTA+0.002%CHX,F组为单独用药组,G组为阴性对照组,均作用24 h后,采用MTT法测量各组生物膜的OD 490 值。在无菌羟磷灰石陶瓷片建立粪肠球菌24 h成熟生物膜模型,随机分为EDTA组、CHX组、EDTA+CHX组、阴性对照组,每组分别由相应药物作用24 h,经荧光染色后置于激光共聚焦扫描显微镜(CLSM)下观察粪肠球菌生物膜变化。结果 A—E组OD 490 值显著低于F、G组(P<0.05),A—E组组间两两比较差异无统计学意义(P>0.05)。CLSM图像显示:成熟生物膜细菌密集、层叠,镜下以绿色荧光染色菌体为主;EDTA组于CLSM下可见生物膜结构稀疏,其中细菌密度及细菌数量显著降低,但仍以绿色荧光为主;CHX组生物膜中细菌数量减少,呈现黄色或桔色,部分区域为深红色;EDTA联合CHX组,生物膜中生物膜结构破坏,细菌数量明显减少,以红色荧光染色菌体为主。结论 EDTA对粪肠球菌生物膜具有分散作用,可增强CHX抗生物膜效果,但其作用效果与浓度无关。 Objective To evaluate the effect of ethylenediaminetetraacetic acid disodium salt(EDTA) as a dispersant on antimicrobial efficacy of chlorhexidine(CHX) against Enterococcus faecalis(E.faecalis) biofilms in vitro.Methods The biofilms of E.faecalis were established in vitro using a 96-well plate,and were treated with 1,2,4,8 or 16 minimal inhibitory concentrations of EDTA+0.002% CHX for 24 hours(groups A,B,C,D and E,respectively).In addition,biofilms treated with CHX alone was selected as group F,and negative control as group G.The OD 490 value was measured by MTT.Mature biofilms were formed by 24-hour incubation on hydroxyapatite disks,and then were divided into EDTA group,CHX group,EDTA+CHX group and negative control group.After treatment for 24 hours,the changes in biofilms were observed using laser confocal scanning microscopy(CLSM).Results Compared with groups F and G,the OD 490 value decreased in groups A,B,C,D and E(P<0.05).No significant differences were found among the five groups(P >0.05).CLSM showed that mature biofilms were composed of densely packed bacteria and mainly displayed green fluorescence.In EDTA group,the biofilms had sparse structure with significantly reduced bacterial density and number,but the bacteria mainly showed green fluorescence.In CHX group,the number of bacteria reduced in biofilms,which mainly showed yellow or orange fluorescence with dark red color in some areas.In EDTA+CHX group,the biofilms showed structural damage with decreased bacterial number,and the bacteria were characterized by red fluorescence.Conclusion EDTA has a dispersing effect on E.faecalis biofilms.It can enhance the anti-biofilm activity of CHX,but its efficacy is independent of the concentration.
