摘要
目的:研究黄连温胆汤对2型糖尿病模型鼠空腹血糖、胰岛素(INS)及肠道菌群变化的影响。方法:选择SPF级SD大鼠,先随机分10只为正常组,其余大鼠均以高糖高脂乳剂1 ml/100g灌胃8 w,并每隔7天监测体重和血糖,8 w后,以35 mg/kg的链脲佐菌素(STZ)腹腔注射诱导2型糖尿病模型,5 d后,测量空腹血糖,选取空腹血糖值大于11.10 mmol/L造模成功的大鼠随机分为:模型组、二甲双胍0.25 g/kg组和黄连温胆汤12 g/kg、6 g/kg、3 g/kg组。各组分别给予等体积相应药物或生理盐水,1次/d,用药4 w后,取材检测。ELISA检测大鼠血清胰岛素的含量;16S-rDNA高通量测序技术对大鼠粪便肠道菌群进行基因测序。结果:与正常对照组比较,模型组空腹血糖明显升高,INS含量明显降低,拟杆菌门(Bacteroidetes)、拟杆菌门+变形菌门(Bacteroidetes+Proteobacteria)、拟杆菌纲(Bacteroidia)占比、拟杆菌门/厚壁菌门比值增加,厚壁菌门(Firmicutes)、梭菌纲(Clostridia)、芽孢杆菌纲(Bacilli)占比降低,sobs、chao、ace、shannon指数降低。黄连温胆汤能使模型鼠空腹血糖值显著降低;血清中INS含量显著升高;肠道菌群中Firmicutes升高,Bacteroidetes、Bacteroidetes+Proteobacteria占比和Bacteroidetes/Firmicutes比值均明显下降,G^+菌Clostridia、Bacilli升高,G^-菌Bacteroidia下降,sobs指数、chao指数、ace指数均明显升高。结论:黄连温胆汤通过调控肠道菌群、提高胰岛素含量、降低血糖,从而达到防治2型糖尿病。
Objective:To study the effect of Huanglian Wendan Tang on FBS,insulin(INS) and gut microbiota in rats with T2D.Methods:SPF rats were selected, 10 in normal group(A), the other rats were fed with high-sugar and high-fat emulsion 1 ml/100 g/d for 8 weeks, their body weights and blood sugar were monitored every seven days. After 8 w, T2D model was established by intraperitoneal injection of 35 mg/kg STZ. Five days later, fasting blood sugar was measured, FBS value more than 11.10 mmol/L were selected and randomly divided into five groups: the model group(B),metformin(0.25 g/kg)group(C) and three Huanglian Wendan Tang groups(12 g/kg,6 g/kg,3 g/kg). After the establishment of the model of T2D, the rats in A, B groups were given normal saline(12 ml/kg), all rats were given corresponding drugs or saline once a day. After 4 weeks of medication, samples were taken for detection. The serum content of INS was detected by ELISA, and 16S-rDNA high-throughput sequencing technology was used to sequence the faecal gut microbiota of rats. Results:Compared with the control group, FBS was increased in the model group, the content of INS was decreased, the proportions of Bacteroidetes, Bacteroidetes+Proteobacteria and Bacteroidia were increased, the ratio of Bacteroidetes/Firmicutes was increased, the proportions of Firmicutes, Clostridia and Bacilli were decreased, the indexes of sobs、chao、ace、shannon were reduced. Compared with the model group, Huanglian Wendan Tang significantly reduced FBS in T2D model rats, the content of INS was significantly increased, Firmicutes was increased, the proportions of Bacteroidetes and Bacteroidetes+Proteobacteria were decreased, the ratio of Bacteroidetes/Firmicutes was decreased, Clostridia and Bacilli of G+ bacteria was increased, Bacteroidia of G-bacteria was decreased, sobs index, Chao index and ace index were increased.Conclusions:Huanglian Wendan Tang can regulate gut microbiota, increase the content of INS and reduce FBS, which may effectively prevent and treat T2D.
