基于网络药理学的黄芪多组分协同治疗乳腺癌的作用机制研究 被引量:4

摘要 运用网络药理学方法,通过TCMSP数据库检索获得黄芪活性化合物,使用OMIM和TTD数据库筛选乳腺癌疾病靶点,利用Cytoscape3.2.1软件构建化合物-疾病靶点网络图,筛选出核心靶点,运用Metascape在线分析平台进行通路富集分析。通过筛选得到与疾病靶点有关的黄芪活性化合物共20个,其主要通过调控AR、ESR1、ESR2、PIK3CG、BCL2等靶蛋白及内分泌阻力、EGFR酪氨酸激酶抑制剂耐药性、凋亡信号通路、PI3K/Akt信号通路等来发挥治疗乳腺癌作用。本研究通过网络药理学初步揭示了黄芪治疗乳腺癌的潜在调控机制,体现了中药多成分、多靶点、多通路协同作用的特点,为黄芪药理研究及临床应用提供理论依据。 Using a network pharmacology method,we retrieved active compounds of Astragalus membranaceus through TCMSP database,screened the targets of breast cancer through OMIM and TTD database,and constructed compound-disease target network diagram adopting Cytoscape 3.2.1 software.Furthermore,we screened core targets,and analyzed pathway enrichment through Metascape online analysis platform.20 Active compounds of Astragalus membranaceus related to disease targets have been obtained,and they play therapeutic roles for breast cancer through mainly regulating target proteins such as AR,ESR1,ESR2,PIK3CG,and BCL2,endocrine resistance,EGFR tyrosine kinase inhibitors resistance,apoptosis signaling pathways,and PI3K/Akt signaling pathways.This study preliminarily reveals the potential regulation mechanism of Astragalus membranaceus therapy for breast cancer through network pharmacology,reflects the characteristics of multi-component,multi-target and multi-pathway of traditional Chinese medicine,and provides a theoretical basis for pharmacological research and clinical application of Astragalus membranaceus.
