泼尼松对小鼠胸腺、脾脏、肝脏脏器指数及肝脏形态学的影响 被引量:11

摘要 目的探讨泼尼松对小鼠胸腺、脾脏、肝脏的脏器指数以及肝脏组织形态学影响,了解泼尼松对小鼠免疫系统及肝脏的影响。方法将雄性昆明小鼠30只随机分成3组:正常组、泼尼松低剂量组(1.8mg/kg)、泼尼松高剂量组(18mg/kg),每组10只。每天用不同剂量泼尼松对小鼠进行灌胃给药,连续给药20d,每隔1天监测小鼠体质量。第21天剥取小鼠的胸腺、脾脏、肝脏,先称重然后计算脏器指数,取肝脏组织做HE染色实验。结果与正常组比较,泼尼松处理组小鼠体质量明显降低(P<0.01),胸腺、脾脏以及肝脏指数显著降低(P<0.01),泼尼松高剂量组下降更明显。肝脏切片HE染色结果显示,与正常组比较,泼尼松处理组细胞核固缩,肝小叶之间出现明显间隙。结论泼尼松对小鼠免疫系统有较强的抑制性,可导致肝脏组织发生病变、萎缩。 Objective To investigate the effects of prednisone on thymus,spleen and liver of mice,and to understand the effects of prednisone on immune system and liver of mice. Methods 30 male KM mice were divided into three groups:normal group,prednisone low-dose group(1.8 mg/kg),and prednisone high-dose group(18 mg/kg),10 in each group. Each treatment group was administered with different doses of prednisone by gavage daily for 20 days.Meanwhile,the body mass of the mice was monitored every other day.At the 21st day,the thymus,spleen,and liver were taken from these mice and weighed to calculate the organ index.And the liver tissue was analyzed by hematoxylin and eosin(HE) staining. Results Compared to the normal mice,the body mass of prednisone groups decreased obviously( P <0.01),the organ index of thymus,spleen,and liver in prednisone groups were remarkably reduced( P <0.01) and the high-dose group showed a more significant decrease.The results of HE staining of liver sections showed that,compared with the normal group,the prednisone group had nuclear condensation and obvious interlobular space. Conclusion Prednisone inhibited the immune system of mice,and may cause the liver tissue to become diseased and atrophied.
