摘要
云南省保存着丰富的马铃薯品种资源,本研究于2016-2017年在云南省采集79份马铃薯品种,以已知的24个主栽品种为对照,进行主要农艺性状的比较;以前期筛选的多态性较好的3对SSR引物和2对细胞质DNA引物,对待测品种进行品种鉴定、遗传多样性分析和构建DNA指纹图谱。结果表明,79份待测品种农艺性状多样化显著且能明显区分出野生型和普通栽培型特点;采集品种中细胞质DNA多态性丰富,具有W/α、W/β、T/α、T/β和W/γ五种类型,野生型比率占63.29%;利用3对SSR引物共扩增出29个等位位点,全部等位位点都具有多态性,多态性比率为100%,构建了待测品种的DNA分子指纹图谱,共鉴定出37个采集品种。结果还发现,79个采集品种间遗传多样性较丰富。
There are abundant potato variety resources conserved in Yunnan Province.In this study,79 copies of potato varieties were collected in Yunnan Province during 2016 to 2017,using the known 24 main cultivated varieties,the main agronomic traits were compared.3 pairs of SSR primers and 2 pairs of cytoplasmic DNA primers with high polymorphisms were screened in the early stage to conduct variety identification,genetic diversity analysis and DNA fingerprint construction of the varieties to be tested.The results showed that the agronomic characters of 79 copies of varieties to be tested had significant diversity and could clearly distinguish the characteristics of wild type from common cultivated type.Cytoplasmic DNA polymorphism was rich in the collected varieties,including 5 types:W/α,W/β,T/α,T/βand W/γ,and the ratio of wild type was 63.29%.A total of 29 allelic loci were amplified by 3 pairs of SSR primers,all allelic loci were with polymorphism(100%),DNA molecular fingerprint of varieties to be tested was constructed and 37 collected varieties were identified in this study.The result also found that the genetic diversity was quite rich among 79 collected varieties.
