摘要
目的研究中药五倍子的主要有效成分五倍子酸对^60 Co-γ射线诱导的小鼠造血系统损伤的防护作用,并对其作用机制进行初步探讨。方法实验动物选用ICR小鼠和BALB/c小鼠。分为正常对照组、辐照组、五倍子酸低剂量组(100 mg/kg GA+辐照)、五倍子酸高剂量组(200 mg/kg GA+辐照)和阳性对照组(WR-2721+辐照)共5组。观察五倍子酸对电离辐射诱导的小鼠脾重、脾系数、脾结节形成能力、外周血中WBC、RBC、LYM、PLT数目以及脾脏中凋亡相关蛋白Bax、Bcl-2、P53含量和凋亡相关激酶Caspase3变化的影响。结果100 mg/kg和200 mg/kg的五倍子酸能显著减轻辐照所致小鼠脾重下降的程度(P<0.05),200 mg/kg的五倍子酸能显著改善脾系数下降的程度,并能明显提高受照小鼠脾结节形成能力(P<0.05)。100 mg/kg和200 mg/kg的五倍子酸能抑制小鼠外周血中RBC、WBC下降的程度,并能明显改善辐射诱导的PLT含量减少(P<0.05)。100 mg/kg和200 mg/kg的五倍子酸能显著降低小鼠脾脏细胞Bax/Bcl-2的比值(P<0.05),但并没有引起p53和Caspase3水平显著下降(P>0.05)。结论五倍子酸对辐射所致小鼠损伤具有一定防护作用,其作用机制可能与保护造血系统和减轻细胞凋亡有关。
Objective To investigate protective effects of gallic acid on ^60Co-γray radiation induced hematopoietic system injuries of mice.Methods ICR mice and BALB/c mice,divided into 5 groups:control group,ray radiation group,100 mg/kg GA+γray radiation group,200 mg/kg GA+γray radiation group,WR-2721+γray radiation group.Observed the spleen weights,spleen indexes,CFU-spleen,RBC,WBC,LYM,PLT counts in the peripheral blood of mice and the expression level of Bax,Bcl-2,P53 and Caspase3 in spleen of mice were observed too.Results GA(100 mg/kg,200 mg/kg)could decreased the injury of the spleen weights,GA(200 mg/kg)could increased the spleen indexe and colony forming units induced by 60Co-γray irradiation compared with the control group significantly(P<0.05).GA(100 mg/kg,200 mg/kg)could weakend the the injury of RBC,WBC,PLT counts in blood induced by 60Co-γray irradiation significantly(P<0.05).GA(100 mg/kg,200 mg/kg)could reduced the rate of Bax/Bcl-2 significantly(P<0.05),but have no significant on the expression of p53 and Caspase3(P>0.05).Conclusion GA has protective effect on 60Co-γray radiation induced mice injuries;the mechanism may be related to the reduction of hematopoietic system injury and cell apoptosis induced by irradiation.
