摘要
目的比较研究赤芍、白芍及芍药苷、芍药内酯苷对急性血瘀证大鼠血液流变学和血管内皮功能的影响,探讨赤芍、白芍对血瘀证作用的物质基础。方法大鼠皮下注射盐酸肾上腺素复合冰水浴复制大鼠急性血瘀证动物模型,以丹参酮胶囊为对照药,通过检测大鼠全血黏度、红细胞最大凝集指数(Red blood cell maximum agglutination index,MAXD)、红细胞最大变形指数(Red blood cell maximum deformation index,MAXDI)水平,用放射免疫法测定血浆内皮素1(endothelin,ET-1)含量、采用比色法测定血清一氧化氮(nitric oxide,NO)含量。观察不同剂量的赤芍、白芍对大鼠全血黏度、MAXD、MAXDI、血清NO、血浆ET-1的影响。采用同样的方法通过检测大鼠全血黏度、血清NO、血浆ET-1指标研究芍药苷、芍药内酯苷对急性血瘀证大鼠的影响。结果与模型组相比,赤芍高剂量组及芍药苷高剂量组、中剂量组各切变率的全血黏度显著降低(P<0.05,P<0.001,P<0.01);赤芍中剂量组、赤芍低剂量组及白芍高剂量组中切变率及低切变率全血黏度降低(P<0.05)。与模型组比较,赤芍高剂量组MAXD、MAXDI均有显著改变(P<0.001,P<0.01),白芍高剂量组和白芍中剂量组MAXD降低(P<0.01)。与模型组比较,赤芍、白芍、芍药苷各剂量组及芍药内酯苷高剂量组、中剂量组血清NO值显著升高(P<0.05);各组血浆ET-1无显著差异。结论赤芍、白芍均能降低急性血瘀证大鼠的血黏度,芍药苷是两者起作用的共同活性成分,芍药内酯苷对血瘀证的作用不及芍药苷;赤芍对血瘀证的作用优于白芍,这与其芍药苷含量较多,而芍药苷有较强改善血液流变学异常和保护血管内皮功能作用有关。
Objective To compare the effects of Radix Paeoniae Rubra,Radix Paeoniae Alba,paeoniflorin and albiflorin on hemorheology and vascular endothelial function in rats with acute blood stasis syndrome.And to explore the functional material basis of Radix Paeoniae Rubra and Radix Paeoniae Alba in blood stasis syndrome.Methods The rat model was replicated by subcutaneous injection of epinephrine hydrochloride combined with ice bath.The control drug was tanshinone capsules.Through testing rats’whole blood viscosity,red blood cell maximum agglutination index(MAXD),and red blood cell maximum deformation index(MAXDI),as well as testing plasma endothelin-1(ET-1)by radioimmunoassay and serum nitric oxide(NO)by colorimetry,the effects on the whole blood viscosity,MAXD,MAXDI,serum NO and plasma ET-1 of rats with different doses of Radix Paeoniae Rubra and Radix Paeoniae Alba were observed.Using the same method,the effects of paeoniflorin and albiflorin on the whole blood viscosity,serum NO and plasma ET-1 of rats with acute blood stasis syndrome were detected.Results Compared with the model group,the whole blood viscosity of each shear rate in the Radix Paeoniae Rubra high-dose group,the paeoniflorin high-dose group and the paeoniflorin medium-dose group was decreased significantly(P<0.05,P<0.001,P<0.01);the whole blood viscosities of medium shear rate and low shear rate in the Radix Paeoniae Rubra medium-dose group,the Radix Paeoniae Rubra low-dose group and the Radix Paeoniae Alba high-dose group were decreased(P<0.05).Compared with the model group,the MAXD and MAXDI in the Radix Paeoniae Rubra high-dose group were changed significantly(P<0.001,P<0.01);the MAXD in the Radix Paeoniae Alba high-dose group and Radix Paeoniae Alba medium-dose group was decreased(P<0.01).Compared with the model group,the serum NO values of the groups of Radix Paeoniae Rubra,Radix Paeoniae Alba and paeoniflorin,the albiflorin high-dose group and the albiflorin low-dose group were significantly increased(P<0.05);there was no significant difference in the plasma ET-1 between each group.Conclusion Radix Paeoniae Rubra and Radix Paeoniae Alba can decrease the whole blood viscosity in rats with acute blood stasis syndrome.Paeoniflorin is a common active component of them.The effect of albiflorin on blood stasis syndrome is not as good as that of paeoniflorin.The effect of Radix Paeoniae Rubra on blood stasis syndrome is better than that of Radix Paeoniae Alba,because it contains more paeoniflorin and paeoniflorin has a strong effect on improving hemorheological abnormalities and protecting vascular endothelial function.
