摘要
采用超声、回流提取方法考察不同溶剂及提取温度对虎耳草提取率的影响;采用多巴速率氧化法和DPPH法测定提取物对酪氨酸酶的激活及抗氧化作用。结果表明,虎耳草70%乙醇超声提取物及其石油醚、乙酸乙酯、正丁醇、水溶剂提取物对酪氨酸酶的激活率分别为26.41%,-2.15%,-2.76%, 17.18%和28.92%;对DPPH自由基具有较强的清除作用, IC50值分别为26.81, 21.44, 10.37, 25.60和245.21μg/m L。虎耳草可作为天然酪氨酸酶激活剂和抗氧化剂进一步开发研究。
To optimize the extracting conditions of Saxifraga stolonifera Meerb, the antioxidant and tyrosinase activities of the extracts were discussed. The extraction conditions were screened by measuring the oxidation rate of DL-dopa. The antioxidation was measured by DPPH scavenging activity. The results showed that the 70% ethanol ultrasonic extract and it’s petroleum ether, ethyl acetate, n-butyl alcohol, water layer parts were active fractions. The activation rates of them to tyrosinase were 26.41%,-2.15%,-2.76%, 17.18%, and 28.92%;The IC50 were 26.81, 21.44, 10.37, 25.60, and 245.21 μg·mL-1 in DPPH test, respectively. The extracts from Saxifraga stolonifera Meerb. showed obvious activating effect on tyrosinase and strong antioxidant properties, and had potential applications in many industries such as food preservation and medical cosmetology.
