桑葚花青素乙醇浸提工艺优化研究 被引量:11

摘要 以桑果干为原料,采用乙醇溶剂浸取法提取桑葚中的花青素,采用pH示差法测定花青素含量,探究料液比、乙醇浓度、提取温度、提取时间和pH对花青素提取量的影响,通过单因素试验和正交试验设计优化乙醇溶剂浸取工艺,并通过验证试验分析得到最佳提取工艺条件。最佳提取工艺条件为:乙醇体积分数为55%,料液比为1∶10(g/mL),提取温度为60℃,提取时间为120 min,溶剂pH为4.0,得到桑葚花青素提取量为2.53 mg/g。乙醇溶剂浸取法简便易行,设备投入少,成本低,适用于桑葚花青素的提取。 Using dried mulberry as the raw material,the anthocyanins in mulberry are extracted by ethanol solvent extraction method,the anthocyanins content is determined by pH differential method,and the effects of material-liquid ratio,ethanol concentration,extraction temperature,extraction time and pH on anthocyanins extraction amount are explored.The ethanol solvent extraction process is optimized by single factor test and orthogonal test design,and the best extraction process is obtained by validation test analysis.The optimum extraction conditions are as follows:the volume fraction of ethanol is 55%,the solid-liquid ratio is 1∶10(g/mL),the extraction temperature is 60℃,the extraction time is 120 min,the solvent pH is 4.0,and the extraction amount of mulberry anthocyanins is 2.53 mg/g.Ethanol solvent extraction method is simple and easy,with less equipment investment,low cost,and it is suitable for the extraction of mulberry anthocyanins.
