摘要 [目的]建立黑叶观音莲组培快繁技术体系。[方法]以黑叶观音莲的顶芽作为外植体,通过对外植体的消毒时间、不定芽诱导、增殖继代和生根培养等环节技术探究,建立黑叶观音莲组培快繁技术体系。[结果]最佳的外植体消毒时间为19 min,污染率为5.6%,褐变率为4.4%;培养基MS+6-BA 5.00 mg/L+NAA 0.20 mg/L最适合不定芽的诱导,诱导率62.2%;最佳的丛生芽增殖培养基为MS+6-BA 3.50 mg/L+NAA 0.30 mg/L,增殖倍数为4.3;最佳的生根培养基为1/2MS+NAA 0.20 mg/L+0.50 g/L活性炭,生根率达100%。[结论]该研究建立了黑叶观音莲组培快繁技术体系,为其工厂化生产提供了理论依据。 [Objective]To establish the tissue culture regeneration system of Acanthopanax amazonica.[Method]With the top bud of the A.amazonica as the explant,the tissue culture regeneration system was established by the disinfection time,adventitious bud induction,proliferation and rooting culture.[Result]The best explant disinfection time was 19 min,the contamination rate was 5.6%,and the browning rate was 4.4%.MS+6-BA 5.00 mg/L+NAA 0.20 mg/L was suitable for adventitious buds in the medium of A.amazonica,the induction rate was 62.2%;MS+6-BA 3.50 mg/L+NAA 0.20 mg/L was suitable for bud proliferation,the proliferation coefficient was 4.3;1/2MS+NAA 0.20 mg/L+0.50 g/L AC was suitable for rooting,the rooting rate reached 100%.[Conclusion]This study established a tissue culture regeneration system of A.amazonica,which provided a theoretical basis for its factory production.
