摘要
目的优选和降颗粒中干姜、枳实混合挥发油的β-环糊精(β-CD)包合工艺,对包合前后混合挥发油的化学成分进行分析。方法采用饱和水溶液法制备包合物,以综合评分为指标通过正交试验优化挥发油包合工艺,通过扫描电镜对包合物进行表征,通过GC-MS对包合前后挥发油成分进行分析。结果最佳包合工艺为挥发油(mL)与β-CD(g)的配比为1∶10,β-CD(g)与水(m L)的配比为1∶20,包合温度40℃,包合3.5 h;扫描电镜,X射线衍射及差示扫描量热分析结果表明混合挥发油包合物已形成;通过GC-MS鉴定出混合挥发油中46种化学成分。结论优选得到的挥发油包合工艺稳定可行,可用于和降颗粒生产,减少了颗粒制备中挥发油的损失,适用于含挥发油固体制剂的生产。
Objective To optimize the process of β-cyclodextrin(β-CD) inclusion compound of the volatile oil in dried ginger and bitter orange in Hejiang granules, and to analyze the chemical composition of volatile oil before and after the inclusion. Methods The inclusion compound was prepared by saturated aqueous solution. The orthogonal test was used to determine the inclusion rate of volatile oil as an index. The inclusion compounds were characterized by SEM. The GC-MS was used to analyze the compositions of volatile oil before and after the inclusion. Results The ratio of volatile oil(mL) to β-CD(g) was 1∶10, the inclusion temperature was 40 ℃, the inclusion time was 3.5 h, and the ratio of β-CD(g) to water(mL) was 1∶20. The results of SEM, XRD and DSC proved that the inclusion compound of volatile oil was formed. Totally 46 chemical constituents in the volatile oil were identified by GC-MS. Conclusion The optimized inclusion of volatile oil is stable and feasible, which can reduce the loss of volatile oil in the granule preparation and is suitable for the production of solid preparation containing volatile oil. The inclusion process can be used in the production of Hejiang granules.
