摘要
目的建立高效液相色谱法测定碘化钾滴眼液中羟苯乙酯的含量。方法色谱柱为Agilent Eclipse Plus C18色谱柱(4.6 mm×250 mm,5μm),流动相为0.005 mol·L^-1醋酸铵溶液[含三乙胺1%,用冰醋酸调节pH值至(5.0±0.5)]-乙腈(50∶50,v/v),检测波长为256 nm,流速为1.0 mL·min^-1,柱温为30℃。结果羟苯乙酯在6.010 0~21.034 9μg·mL^-1浓度范围内呈良好线性关系,r=0.999 9,平均回收率为99.83%,RSD=0.70%(n=9),该色谱方法重现性好,对照品溶液和供试品溶液在室温放置24 h基本稳定。结论该方法简单、快速、准确,可用于测定碘化钾滴眼液中羟苯乙酯的含量。
OBJECTIVE To establish an HPLC method for the determination of ethylparaben in potassium iodide eye drops.METHODS The analytic column was Agilent Eclipse Plus C18 (4.6 mm×250 mm,5 μm) and the mobile phase was 0.005 mol·L^-1 ammonium acetate (1 000 mL consisted of 10 mL triethanolamine,p H adjust to 5.0 with acetic)-acetonitrile (50 ∶50,v/v) at a flow rate of 1.0 mL·min^-1. The detection wavelength was 256 nm and the column temperature was 30 ℃ . RESULTS The linear range of ethylparaben was 6.010 0-21.034 9 μg·mL^-1,r= 0.999 9.The average recovery rate of ethylparaben was 99.83% with RSD= 0.70% (n= 9).The method had high reproducibility. The reference solution and testing solution were stable for 24 h in room. CONCLUSION The detection method is simple,quick and accurate,which is suitable for the determination of ethylparaben in potassium iodide eye drops.
