摘要
稻瘟病是导致粮食减产的重要病害之一。开发抗稻瘟病基因Pi1的分子标记并用于抗病分子育种是防治稻瘟病危害的有效途径。本研究根据Pi1与Piks、Pikh、Pikm、Pikp、Pik、Pi7基因序列之间的SNP开发了荧光分子标记MM-Pi1,并利用其对94份水稻亲本材料进行检测,结果发现‘安丰B’与‘96B’中含有Pi1基因,52份水稻亲本含有与Pi1间具1个碱基差异的等位基因,40份水稻亲本中该等位基因缺失。对其中的10份水稻亲本材料的等位基因进行测序分析,进一步验证了分子标记检测结果的准确性。稻瘟病抗性鉴定试验结果表明,包含Pi1基因的‘安丰B’与‘96B’均对稻瘟病有高抗性。因此,Pi1荧光分子标记为水稻抗稻瘟病分子育种提供了新的基因分型方法。
Rice blast is one of the most important diseases to reduce grain yield.Developing molecular marker of rice blast resistance gene Pi1 and using it in molecular breeding of disease resistance is an effective approach to reduce the damage caused by rice blast.In this study,the fluorescent molecular marker of Pi1 MM-Pi1 was developed on the basis of the SNP differences between Pi1 and Piks,Pikh,Pikm,Pikp,Pik,Pi7.94 rice parental materials were tested with the marker,and the results showed that ’AnfengB’ and ’96 B’ have Pi1 gene,52 rice parental materials have its allele with one nucleotide difference,and 40 rice parental materials have deletion allele.The allele of 10 rice parent materials were sequenced to further confirm that the molecular marker testing result was reliable.Further rice blast inoculation result showed that both ’AnfengB’ and ’96 B’ are resistant to rice blast disease.Therefore,MM-Pi1 developed in this study provides a novel functional marker for rice disease resistance breeding.
