摘要
目的观察麝香保心丸对代谢综合征(MS)大鼠心肌组织尾加压素Ⅱ(UⅡ)及其受体(UT)的影响。方法取雄性SD大鼠24只,先随机分成正常对照组(8只)和MS组(16只),分别用普通饲料和高果糖饲料喂养,用高果糖饮食诱导MS大鼠模型后,随机分为模型对照组和麝香保心丸组,取大鼠心肌组织应用Masson染色观察血管胶原沉积情况,比较心肌肥厚指数,应用放射免疫法检测大鼠血浆和心肌组织UⅡ含量,应用免疫组化法检测心肌组织UT表达。结果高果糖饮食可成功诱导典型MS大鼠模型,与正常对照组相比,Masson染色可见MS组大鼠心肌组织有明显胶原沉积,心肌肥厚指数增加,UⅡ/UT表达亦同步上调。用麝香保心丸干预后,心肌组织胶原沉积和心肌肥厚指数改善,UⅡ/UT表达下调。结论麝香保心丸可下调UⅡ/UT表达,改善心肌重构。
Objective To investigate the effect of Shexiang Baoxin Pill(SXBXP)on myocardial tissue tail urotensinⅡand its receptor in rats with metabolic syndrome(MS).Methods After inducing an MS rat model with a high-fructose diet,the rats were randomly divided into model control group and SXBXP group.The expression of collagen was determined by Masson staining and myocardial hypertrophy index was compared.The urotensinⅡcontents were detected by radioimmunoassay,and the expressions of urotensinⅡreceptor were determined by immunohistochemistry.Results The MS model of rats were induced by high-fructose diet successfully.Compared with normal control group,Masson staining showed that there were collagen deposited in myocardial tissue of MS group,and myocardial hypertrophy index was increased.The expression of urotensinⅡand its receptor also increase simultaneously.Conclusion Shexiang Baoxin Pill could reduce the expression of urotensinⅡand its receptor,and improve the myocardial remodeling.
