摘要 副干酪乳杆菌(Lactobacillus paracasei)作为乳制品中常见的重要益生菌,能够增强机体免疫力、改善调节肠道菌群平衡、降低血糖水平等。黏附是微生物与宿主相互作用的先决条件之一,是其发挥生物屏障功能的基础。L.paracasei作为益生菌吸附于黏膜表面可以延长自身作用于宿主的时间。基于基因组学分析L.paracasei的黏附作用及其对病原菌黏附的抑制作用,有助于进一步探讨益生菌的生物屏障机制,对乳酸菌的研究开发具有非常重要的理论意义。本课题以分离自西藏地区传统发酵奶制品的L.paracasei为研究对象,筛选出黏附作用最显著的L.paracasei BS 08,分析其黏附作用相关基因,通过去除菌体表面的黏附素,观察对Caco-2细胞的黏附差异性,并探究目标菌株对病原菌黏附的抑制作用。 As a common and important probiotic in dairy products,Lactobacillus paracasei can enhance the body's immunity,improve the bal⁃ance of intestinal flora,reduce blood sugar level,etc.Adhesion is one of the prerequisites for interaction between microorganisms and their hosts.Adhesion is also the basis for their function as biological barriers.Adsorption of L.paracasei as probiotics on the mucosal surface can pro⁃long the time of its own action on the host.Adhesion of L.paracasei based on genomics analysis and its inhibition on pathogen adhesion are helpful to explore the biological barrier mechanism of probiotics and have great theoretical significance for the research and development of Lactic acid bacteria.In this research,the L.paracasei isolated from the traditional fermented dairy products of Tibet.L.paracasei BS 08 with the best adhesive abilities were selected,the adhesion-related genes were analyzed.Differences of adhesion to Caco-2 cells were observed by re⁃moving the adhesion element on the surface of the bacteria.The inhibition effect of the target strain on the adhesion of pathogenic bacteria was investigated.
