摘要
【目的】为了培育炸荚抗性新品种,提高大豆产量。【方法】利用前期通过同源克隆方法得到的大豆GmAGL8基因构建的pCAMBIA1300∷GmAGL8和pCAMBIA1300∷antiGmAGL8载体,采用花浸法对野生型拟南芥(Arabidopsis thaliana)进行遗传转化,对转基因拟南芥进行了PCR鉴定、表型观察与分析。【结果】对比野生型,过表达GmAGL8的转基因拟南芥叶片相对较厚,呈圆形,出现早花现象,果荚成熟提前,果荚成熟后2周少量果荚出现开裂;对比野生型反义抑制GmAGL8植株叶片较薄呈长圆形,开花较迟,果荚成熟晚且短小,角果成熟时果荚完全炸开。【结论】GmAGL8对拟南芥叶片发育,开花时间以及果荚开裂性状均有影响,在拟南芥中过表达GmAGL8基因可在一定程度上提高果荚开裂抗性。
【Objective】 The pod-shattering habit has a serious effect on soybean production, thus the research of soybean pod-shattering is of great significance for breeding new varieties of pod-shattering resistance and increasing the soybean yield. 【Methods】 We cloned a gene named GmAGL8 which had deep homology with Arabidopsis FUL and peach PpMADS6 genes from the soybean genome by the method of homologous cloning. In this study, we constructed two vectors, pCAMBIA1300::GmAGL8 and pCAMBIA1300::antiGmAGL8, then transferred these two vectors to wild-type of Arabidopsis thaliana by Agrobacterium flower dipping. PCR detection, phenotype observation and analysis of transgenic Arabidopsis thaliana were carried out. 【Results】 The leaves of transgenic Arabidopsis thaliana which overexpressed GmAGL8 were relatively thick and round, showing early flowering, pod ripened earlier and the pod rarely cracked two weeks after ripening compared to the wild type. Antisense-suppressing GmAGL8 plant leaves were oblong, flowering later, fruit pods matured late and short, and when the carobs matured, the fruit pods completely exploded. 【Conclusions】 GmAGL8 has effects on leaf development, flowering time and time of pod-shattering. Overexpression of GmAGL8 in Arabidopsis can inhibit flowering and improve pod-shattering resistance.
