母鸡蛋清混样检测禽白血病病毒p27抗原的应用研究 被引量:4

Detection of avian leukosis virus antigen p27 using mixed albumen

摘要 为探讨同一母鸡蛋清混样法检测禽白血病病毒p27抗原用于禽白血病净化的可行性,本研究对3个蛋清逐一检测得到的43只阳性鸡和49只阴性鸡,再次进行了混样检测,通过对比2种方法的判定结果,发现混样检测法需结合样品与阳性对照OD值的比值(S/P)≥0.10的阳性判定标准才能够检出所有阳性个体,此外降低S/P阈值会有助于将一部分弱阳性或可疑个体检出。进一步采用蛋清混样检测法对北京油鸡4个品系开展了2~3次的净化流程后,发现1个品系的阳性比例没有变化,而另外3个品系阳性比例持续降低,并且有一个品系(L2系)首次实现了零检出。对4个品系808只公鸡进行了2次全血病毒分离,结果显示阳性率均为0,可疑(S/P≥0.10)比例均低于2%,净化效果显著。本结果充分证实了蛋清混样检测结合S/P≥0.10的阳性标准开展禽白血病净化是行之有效的,为其他品种禽白血病的净化提供参考。 In order to study the feasibility of detecting avian leukosis virus(ALV)p27 antigen for AL eradication using mixed albumen from several eggs of one hen,43 positive chickens and 49 negative ones obtained from a single albumen detection were selected and tested again using a mixed albumen detection.Comparison of the results revealed that only by combining the positive criterion of S/P ratio(the ratio of OD value between the sample and the positive control)≥0.10 could all the positive individuals be covered with the mixed albumen method.At the same time,decreasing the S/P threshold value contributed to eliminating some weakly positive or suspicious birds.Furthermore,the mixed albumen detection method was applied for AL eradication in four Beijing You chicken lines.After two or three times of detection,the positive rate of one line did not change,while those of the other 3 lines dropped continuously and quickly,among which line L2 had a detection result of zero.Meanwhile,808 male birds from the 4 lines were detected twice using the method of virus isolation from the whole blood.No positive males were observed and even the percentage of suspicious ones(S/P≥0.10)was below 2%,indicating an excellent eradicating result.Our findings well suggested that,in combination of the positive criterion of S/P≥0.10,the mixed albumen detection method was effective for AL eradication,which provided valuable and referable information for AL eradication in other chicken breeds.
