七叶皂苷钠对宫颈癌模型大鼠癌细胞凋亡的影响及作用机制研究 被引量:5

摘要 目的研究七叶皂苷钠对宫颈癌模型大鼠癌细胞凋亡的影响及作用机制。方法选取清洁级健康雄性SD大鼠30只,建立宫颈癌模型,分为模型组、七叶皂苷钠组和正常组,每组10只。计算大鼠肿瘤体积、胸腺、脾脏指数,进行病理学观察、计算宫颈癌细胞增值率、凋亡率,检测Caspase-3、Caspase-7、Caspase-9、PI3K、pAKT蛋白。结果与正常组比较,模型组和七叶皂苷钠组大鼠肿瘤体积增加,胸腺指数、脾指数降低(P<0.05);与模型组比较,七叶皂苷钠组肿瘤大鼠体积增加,胸腺指数、脾指数降低(P<0.05)。与正常组比较,模型组和七叶皂苷钠组大鼠癌细胞增殖率、凋亡率高(P<0.05);与模型组比较,七叶皂苷钠组大鼠癌细胞增殖率降低,凋亡率升高(P<0.05)。与正常组比较,模型组和七叶皂苷钠组大鼠Caspase-3、Caspase-7、Caspase-9蛋白表达降低(P<0.05);与模型组比较,七叶皂苷钠组大鼠Caspase-3、Caspase-7、Caspase-9蛋白表达升高(P<0.05)。与正常组比较,模型组和七叶皂苷钠组大鼠PI3K、pAKT蛋白表达升高(P<0.05);与模型组比较,七叶皂苷钠组大鼠PI3K、pAKT蛋白表达降低(P<0.05)。结论七叶皂苷钠能够减小宫颈癌大鼠肿瘤体积,改善胸腺指数、脾指数,通过激活PI3K、pAKT信号通路,调控Caspase-3、Caspase-7、Caspase-9蛋白表达,起到降低癌细胞增殖,促进凋亡作用。 Objective To investigate the effects of sodium aescinate on apoptosis of cervical cancer cells in rats,and to explore its action mechanism.Methods Thirty healthy clean class male SD rats were were enrolled in the study,and the animal models with cervical cancer were established.The experiment was divided into model group,sodium aescinate group and control group,with 10 rats in each group.The tumor’s size,thymus and spleen indexes were calculated.And pathological observation was performed,and cell proliferation rate and apoptosis rate of cervical cancer were calculated.Moreover the levels of Caspase-3,Caspase-7,Caspase-9,PI3K and pAKT protein were detected and compared among the three groups.Results As compared with those in control group,the tumor’s size in model group and sodium aescinate group was significantly increased,but the thymus and spleen indexes were significantly decreased(P<0.05).As compared with those in control group,the proliferation rate and apoptosis rate of cancer cells in model group and sodium aescinate group were significantly increased(P<0.05).As compared with those in model group,the proliferation rate in sodium aescinate group was significantly decreased,however,apoptosis rate of cancer cells was significantly increased(P<0.05).As compared with those in control group,the expression levels of Caspase-3,Caspase-7 and Caspase-9 in model group and sodium aescinate group were significantly decreased(P<0.05).As compared with those in model group,the expression levels of Caspase-3,Caspase-7 and Caspase-9 protein in sodium aescinate group were significantly increased(P<0.05).As compared with those in control group,the expression levels of PI3K and pAKT protein in model group and sodium aescinate group were significantly increased(P<0.05).As compared with those in model group,the expression levels of PI3K and pAKT protein in sodium aescinate group were significantly decreased(P<0.05).Conclusion Sodium aescinate can decrease the tumor’s size in rats with cervical cancer,improve thymus index and spleen index,regulate the expression levels of Caspase-3,Caspase-7 and Caspase-9 proteins by activating PI3K and pAKT signaling pathways,which plays a role in reducing the proliferation of cancer cells and promoting cell apoptosis.
