摘要
目的:初步探讨淫羊藿素(Icaritin,ICT)是否通过调控NF-κB信号保护心肌缺血再灌注损伤大鼠。方法:采用Real time-PCR和Western Blot方法检测IL-6、IL-1β、TNF-α等炎症因子的mRNA及(或)蛋白表达水平,同时检测NF-κB mRNA及P65蛋白表达水平。结果:经ICT治疗后,心肌组织中IL-6、TNF-α的mRNA水平下降,IL-6、IL-1β的蛋白表达水平也显著下降;并且ICT还可有效降低MIRI大鼠NF-κB mRNA表达水平及P65蛋白表达水平。结论:ICT可能通过抑制NF-κB信号的活化减轻MIRI所诱导的炎症损伤,进而对心肌缺血再灌注损伤大鼠发挥保护作用。
Objective:To investigate the protective effect of Icaritin(ICT)on myocardial ischemia reperfusion injury in rats by regulating NF-κB signaling activity.Methods:Real time-PCR or Western Blot was used to detect the mRNA or protein levels of inflammatory factors such as IL-6,IL-1β,TNF-α,as well as the mRNA expression of NF-κB and the protein expression of P65.Results:Compared with the vehicle group after ICT treatment,the mRNA levels of IL-6,TNF-αin myocardial tissue were significantly decreased,which was consistent with the protein expression of IL-6,IL-1β.It shows that ICT could decrease the level of inflammation in myocardial tissue.It was found out that the mRNA expression of NF-κB and protein expression of P65 were significantly decreased.Conclusion:ICT could reduce the inflammatory damage induced by MIRI via inhibiting the activization of NF-κB signaling,and it also has protective effect on myocardial ischemia-reperfusion injury.
