摘要
目的完善和提高倒扣草质量标准。方法以倒扣草对照药材和齐墩果酸对照批为对照建立薄层鉴别方法;照中国药典2015年版通则方法,测定样品的水分、总灰分、酸不溶性灰分和浸出物含量;采用高效液相色谱(HPLC)法测定药材中β-蜕皮甾酮的含量,色谱柱为Kromasil 100-5C 18(250mm×4.6mm,5μm),乙腈-0.1%甲酸溶液(13∶87,V/V),检测波长为250nm,柱温为30℃,进样量为10μL。结果性状及鉴别方法专属性好,样品的水分、总灰分、酸不溶性灰分、浸出物、β-蜕皮甾酮含量的平均值分别为11.3%、10.5%、0.7%、14.4%、0.021%。结论本研究所建立的方法简便、准确、专属性强,可为提高和完善倒扣草质量标准提供实验依据。
OBJECTIVE To optimize and improve the quality standards of Achyranthes aspera L.METHODS The TLC identification was established using both reference crude herb and oleanolic acid as the reference substances,and the contents of water,total ash,acid-insoluble ash and extractive were carried out according to the methods of Chinese Pharmacopoeia(2015 edition).The content ofβ-ecdysterone was analyzed by HPLC method.The deter mination was performed on Kromasil 100-5C 18 column(250mm×4.6mm,5μm)with mobile phase consisted of acetonitrile-0.1%formic acid solution(13∶87,v/v).The detection wavelength was set at 250nm,and the column temperature was 30℃.The sample size was 10μL.RESULTS The macroscopical identification,microscopic features and TLC method were proper.The average contents of water,total ash,acid-insoluble ash,water-soluble extractive and to the content ofβ-ecdysterone were 11.3%,10.5%,0.7%,14.4%,0.021%,respectively.CONCLUSION The experimental data and the established method could provide the experimental basis for the improvement of the quality standard of Achyranthes aspera L.
