摘要
目的:探讨茯苓提取物对高脂饮食致肠道菌群失调小鼠的影响。方法:昆明种(KM)小鼠按体质量随机分为对照组、模型组和茯苓不同提取物(醇提取、水提取和碱提取部位)处理组。利用高脂饮食制备小鼠肠道菌群失调模型,给予茯苓不同提取物后,检测小鼠体质量及血清中高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、三酰甘油(TG)及总胆固醇(TC)水平;苏木-伊红(HE)染色法检测小鼠肝和肠组织病理学形态;结合16 S rRNA测序,探究茯苓不同提取物对小鼠肠道菌群结构的影响。结果:茯苓提取物可在一定程度减缓高脂饮食诱导的小鼠体质量增加,改善4项血脂指标水平,减轻高脂饮食对肝和肠组织的损伤,并可不同程度降低厚壁菌门(Firmicutes)相对丰度,提高拟杆菌门(Bacteroidetes)相对丰度。结论:茯苓提取物具有改善肥胖、调节血脂代谢及调整肠道菌群结构的作用。
Objective:The aim of this study is to evaluate the effects of Poria cocos extracts on dysbacteria induced by high-fat diet in mice.Methods:A dysbacteria model of mice was prepared by feeding with high-fat diet.The body weight,four lipid indexes[high density liptein cholesterol(HDL-C),low density lipoproteins cholesterol(LDL-C),triacylglycerol(TG)and total cholesterol(TC)],histopathology of liver and intestinal tissue,and gut microbiota of mice were observed after administration of the different P.cocos extracts(including ethyl alcohol extract,water extract and alkaline water extract).Results:All the P.cocos extracts could reduce the weight gain induced by high-fat diet at a certain extent,regulate the levels of four blood lipids(HDL-C,LDL-C,TG and TC),reduce the damages of liver and intestinal tissue,increase the relative abundance of Bacteroidetes while reduce the relative abundance of Firmicutes.Conclusion:The extracts of Poria cocos has the protective effects in resisting high-fat-induced obesity by targeting on the gut microbiota.
