大黄和肉苁蓉水煎液对家兔离体小肠运动影响的研究

Effect of Decoction of Rhubarb and Cistanche on the Movement of Isolated Small Intestine of Rabbits

目的比较大黄及肉苁蓉水煎液对家兔离体小肠平滑肌收缩性的影响。方法取健康成年新西兰家兔十二指肠肠段,随机分为8组,分别放入恒温灌流装置中给予不同干预,正常对照组加入0.2 ml生理盐水;阳性对照组加入0.2 ml浓度为1∶10000的乙酰胆碱;大黄水煎液组加入0.01 g/ml的大黄0.2 ml;肉苁蓉水煎液组加入0.01 g/ml的肉苁蓉0.2 ml;维拉帕米+大黄组先加入维拉帕米0.2 ml,待波形稳定后再加入大黄水煎液0.2 ml;阿托品+大黄组、维拉帕米+肉苁蓉组和阿托品+肉苁蓉组的操作方法同维拉帕米+大黄组。进入VBL-420F系统,记录其收缩张力和频率的变化。结果阳性对照组、大黄水煎液组、肉苁蓉水煎液组对家兔离体小肠平滑肌收缩频率和收缩波幅均大于正常对照组,阿托品和维拉帕米对家兔离体小肠平滑肌的收缩频率和振幅低于正常对照组,差异有统计学意义(P<0.05);加入肉苁蓉水煎液后,阿托品+肉苁蓉水煎液组对家兔离体小肠平滑肌的收缩频率及收缩振幅的影响均开始逐渐增高,差异有统计学意义(P<0.05);但维拉帕米+肉苁蓉水煎液组对家兔离体小肠平滑肌收缩频率和振幅的影响均不明显,差异无统计学意义(P>0.05);阿托品+大黄组和维拉帕米+大黄组家兔离体小肠平滑肌收缩频率和振幅变化与加入前比较,差异无统计学意义(P>0.05)。结论大黄及肉苁蓉对家兔离体小肠平滑肌收缩活动均有促进作用,此影响作用均可被维拉帕米阻断,但阿托品能阻断大黄的促进作用却无法阻断肉苁蓉的促进作用。

Objective To Compare the effects of decoction of rhubarb and cistanche on the contractility of isolated rabbit small intestine smooth muscle.Methods The duodenal intestines of healthy adult New Zealand rabbits were randomly divided into 8 groups,and they were put into a constant temperature perfusion device for different interventions.The normal control group was added with 0.2 ml of normal saline;The positive control group was added with 0.2 ml of acetylcholine with a concentration of 1:10000;the rhubarb decoction group was added with 0.2 ml of 0.01 g/ml of rhubarb;Cistanche cistanche decoction group add 0.01 g/ml cistanche 0.2 ml);Verapamil+Rhubarb group first add 0.2mL of Verapamil,and then add 0.2 ml of Rhubarb decoction after the waveform is stable;The operation method of atropine+rhubarb group,verapamil+cistanche group and atropine+cistanche group is the same as verapamil+rhubarb group.Enter the VBL-420F system and record changes in its contraction tension and frequency.Results The contractile frequency and amplitude of small intestine smooth muscle in positive control group,rhubarb decoction group and Cistanche deserticola Decoction group were higher than those in normal control group,while the contractile frequency and amplitude of small intestine smooth muscle in atropine and verapamil group were lower than those in normal control group(P<0.05)The effects of verapamil and Cistanche deserticola Decoction group on the contraction frequency and amplitude of isolated small intestinal smooth muscle of rabbits were not significant(P>0.05);the effects of atropine and rhubarb group and verapamil and rhubarb group on the contraction frequency and amplitude of isolated small intestinal smooth muscle of rabbits were not significant(P>0.05)There was no significant difference in the contractile frequency and amplitude of intestinal smooth muscle before and after the addition(P>0.05).Conclusion Both rhubarb and Cistanche can promote the contraction of isolated small intestinal smooth muscle of rabbits.The same is that this effect can be blocked by verapamil,The difference is that atropine can block the promotion of rhubarb but cannot block the promotion of Cistanche.
