摘要
目的:建立HPLC法测定治咳川贝枇杷露中苯甲酸、羟苯乙酯的含量。方法:色谱柱采用Agilent Eclipse XDB-C18柱(250 mm×4.6 mm,5μm),流动相:甲醇-0.02 moL·L^(-1)乙酸铵溶液,梯度程序;流速:1.0 mL·min^(-1);检测波长:0~8 min为230 nm,8~20 min为254 nm;进样量:10μL;柱温:25℃。结果:苯甲酸在1.00~5.01μg(r=0.9999)范围内呈现良好的线性关系,羟苯乙酯在0.08~0.41μg(r=1.0000)范围内呈现良好的线性关系,平均加样回收率分别为98.92%(RSD=1.71%,n=9)、100.52%(RSD=1.32%,n=9)。结论:该方法简便准确、专属性强,重现性好,可用于治咳川贝枇杷露中防腐剂的含量测定。
Objective To establish a HPLC method to determine the content of benzoic acid and ethyl hydroxybenzoate in Zhike Chuanbei Pipa syrups.Methods HPLC method was used with Agilent Eclipse XDB-C18 column(250 mm×4.6 mm,5μm).The mobile phase was methanol—0.02 moL·l^(-1) ammonium acetate,gradient elution with a flow rate of 1.0 mL·min^(-1),the detection wavelength was set 230 nm in 0~8 min and 254 nm in 8~20 min,sample volume was 10μL and the column temperature was 25℃.Results The linear range of benzoic acid was 1.00~5.01μg(r=0.9999)and ethyl hydroxybenzoate was 0.08~0.41μg(r=1.0000),the average recovery of benzoic acid was 98.92%(RSD=1.71%,n=9)and ethyl hydroxybenzoate was 100.52%(RSD=1.32%,n=9).Conclusion The method is simple,accurate with good specificity and reproducibility,and it can be used in the quantitative determination of preservative in Zhike Chuanbei Pipa syrups.
