一种新型体外溶血检测技术的建立及应用

摘要 目的:建立一种新型体外溶血检测技术,检测血清中处于低水平的血型抗体.方法:取33份分离后的患者单个核细胞用自体血浆重悬后,与本研究的溶血检测体系反应,通过Hb胶体金试纸条进行定性检测,然后与盐水法和凝胶卡法的间接抗人球蛋白检测结果比较,通过SPSS 19.0非参数检测(Mann-Whitney Test)对数据统计分析.结果:新型溶血技术与盐水法和凝胶卡法的间接抗人球蛋白检测相比,新型溶血技术对不同性质不规则抗体(IgG/IgM型)检出率高于盐水法和凝胶法.针对不同种类(Rh、MNSs、Duffy、Kidd、Lewis及JMH系统和自身抗体)的不规则抗体,该技术检出率高于其他两种方法(P<0.05).结论:新型体外溶血检测技术提高了检测灵敏度,有效检出各种不规则抗体,避免反复输血患者的溶血性输血反应,保障输血安全. Objective:To establish a new in vitro hemolysis detection technology for detecting low level blood group antibodies in serum.Methods:A total of 33 samples of isolated mononuclear cells were resuspended with autologous plasma,and reacted with the hemolysis detection system in this study.The Hb colloidal gold strip was used for qualitative detection,and then the results were compared and analyzed with the results of indirect anti-human globulin detection by saline method and gel card method.Statistical significance was assessed using Mann-Whitney Test in SPSS 19.0.Results:Compared with the indirect anti human globulin test of brine and gel card,the new hemolysis technology had higher detection rate of irregular antibodies(IgG/IgM)than brine and gel method.The detection rate of irregular antibodies of different kinds(RH,MNSs,Duffy,Kidd,Lewis and jmh system and autoantibodies)was higher than that of other two methods(P<0.05).Conclusion:The new in vitro hemolysis detection technology can improve the sensitivity of detection,detect various irregular antibodies effectively,avoid the hemolytic transfusion reaction of patients with repeated blood transfusion,and ensure the safety of blood transfusion.
