摘要
目的探讨与研究白芍总苷Wntβ/catenin信号通路对便秘大鼠胃肠功能的治疗作用及机制。方法48只雄性SD大鼠按照随机数字表法平分为3组-模型组、柳氮磺砒啶组与白芍总苷组,所有大鼠均建立了便秘模型。建模成功后模型组、柳氮磺砒啶组与白芍总苷组分别给予0.9%氯化钠溶液、柳氮磺砒啶400 mg/kg、白芍总苷25 mg/kg灌胃,连续治疗21 d。检测3组大鼠2 h大便数量与湿重,疾病活动指数(DAI)评分与结肠黏膜损伤指数(CMDI)评分,血清白细胞介素(IL)-10与IL-17含量,Wntβ/catenin信号通路蛋白表达变化情况。结果治疗21 d后,柳氮磺砒啶组与白芍总苷组大鼠的2 h大便数量与湿重高于模型组,差异均有统计学意义(P<0.05),白芍总苷组大鼠高于柳氮磺砒啶组,差异均有统计学意义(P<0.05)。治疗21 d后,柳氮磺砒啶组与白芍总苷组大鼠的DAI评分与CMDI评分、血清IL-10与IL-17含量、结肠组织Wnt与β-catenin蛋白相对表达水平均低于模型组,差异均有统计学意义(P<0.05),白芍总苷组大鼠低于柳氮磺砒啶组,差异均有统计学意义(P<0.05)。结论白芍总苷在便秘大鼠的应用能抑制Wntβ/catenin信号通路的激活,并抑制炎性因子的表达,从而改善胃肠功能,缓解便秘症状。
Objective To investigate and study the therapeutic effect and mechanism of Baishaozonggan for Wntβ/catenin on gastrointestinal function in constipated rats.Methods A total of 48 male SD rats were divided into 3 groups according to the random number table method-model group,sulfasalazine group and total glucosides of paeony.All rats were established constipation models.After successful modeling,the model group,the sulfasalazine group and the Baishaozonggan were given normal saline,400 mg/kg of the sulfasalazineand 25 mg/kg of the total glucosides of paeony by gavage treatment respectively,continuous treatment for 21 days.The number of stools and wet weight at 2 h,disease activity index(DAI)score and colonic mucosal damage index(CMDI)score,the levels of serum interleukin(IL)-10 and IL-17,Wntβ/catenin signaling pathway protein in the 3 groups of rats Changes in expression were detected.Results After treatment,the stool volume and wet weight of the sulfasalazine group and the Baishaozonggan group were higher than the model group,the differences were statistically significant(P<0.05),and the Baishaozonggan group were higher than the sulfasalazine group,the differences were statistically significant(P<0.05).After treatment,the DAI score and CMDI score,serum IL-10 and IL-17 levels,and the relative expression levels of Wnt andβ-catenin protein in the colon tissue of the sulfasalazine group and the Baishaozonggan group were lower than those of the model group,the differences were statistically significant(P<0.05),the Baishaozonggan groupwere lower than the sulfasalazine group,the differences were statistically significant(P<0.05).Conclusion The application of Baishaozonggan in rats with constipation can inhibit the activation of Wntβ/catenin signaling pathway,and inhibit the expression of inflammatory factors,thereby improve gastrointestinal function and alleviating symptoms of constipation.
