摘要
为检测石榴花水提物(PEW)的抗炎功效,首先测定PEW对巨噬细胞的体外安全浓度,并在安全浓度下检测PEW对脂多糖(LPS)诱导炎症模型巨噬细胞一氧化氮(NO)、白介素6(IL-6)和肿瘤坏死因子-α(TNF-α)分泌水平的影响,最后通过测定PEW对二甲苯所致的小鼠耳郭肿胀的抑制作用,观察PEW的抗炎作用。结果显示,PEW在10~100μg/mL均能够显著抑制LPS诱导炎症模型巨噬细胞NO的分泌(P<0.05),其中100μg/mL浓度时的抑制作用最强。经LPS诱导后炎症模型巨噬细胞能够分泌大量IL-6及TNF-α,PEW能够有效抑制炎症模型巨噬细胞IL-6及TNF-α的分泌。PEW对二甲苯致小鼠耳郭肿胀的影响试验结果显示,高剂量(500μg/mL)PEW对二甲苯致小鼠耳郭肿胀的抑制效果最佳,抑制率可高达60%,耳肿胀度和阳性药物地塞米松组无显著性差异(P>0.05)。由体内外试验可知,PEW体内外均具有良好的抗炎活性,其中高浓度PEW(500μg/mL)的活性最强。本试验为石榴花进一步开发利用提供试验依据。
To test the anti-inflammatory effect of pomegranate flower water extract(PEW),we first measured the safe concentration of PEW on macrophages in vitro,and then measured the effect of PEW on the secretion of NO,IL-6 and TNF-αby macrophages in lipopolysaccharides(LPS)induced inflammatory model.Finally,we observed the anti-inflammatory effect of PEW by measuring the inhibition of PEW on xylene induced ear swelling in mice.The results showed that PEW could significantly inhibit the NO secretion in LPS-induced macrophages at concentrations of 10-100μg/mL(P<0.05),especially at the concentration of 100μg/mL.After LPS induction,macrophages secreted a lot of IL-6 and TNF-α.PEW effectively inhibited the secretion of IL-6 and TNF-αby macrophages.The effect of PEW on xylene induced auricle swelling in mice showed that high dose PEW(500μg/mL)had the best inhibitory effect,and the inhibition rate was as high as 60%.However,there was no significant difference between the degree of auricle swelling and dexamethasone group(P>0.05).In vitro and in vivo experiments showed that PEW had good anti-inflammatory activity,and the high concentration of PEW(500μg/mL)was the strongest.This study provides experimental basis for further development and utilization of pomegranate flower.
