摘要
目的探讨骨髓来源间充质干细胞(bone marrow-derived mesenchymal stem cells,BMSCs)复合PLGA膜支架对牙周组织再生的影响。方法体外分离培养比格犬BMSCs,CCK8检测BMSCs细胞生长曲线,流式细胞术检测BMSCs细胞纯度;溶剂浇铸法制备PLGA膜,检测PLGA膜的吸水率和溶解率;分别对培养皿单层培养的BMSCs和BMSCs复合PLGA膜培养的细胞行成骨诱导分化,茜素红和ALP染色检测成骨细胞分化率,WB检测细胞Col1、Runx2、osterix蛋白表达;手术制备比格犬牙周组织缺损模型,分别以生理盐水注射组、BMSCs注射组、PLGA膜移植组和BMSCs复合PLGA膜移植组治疗,以无缺损牙作为对照组。结果BMSCs复合PLGA膜培养的细胞增殖水平和成骨诱导分化率显著高于单层培养的BMSCs(P<0.01);BMSCs复合PLGA膜组可促进比格犬牙周组织再生,效果优于其他组。结论PLGA膜可为BMSCs提供良好的微环境,促进细胞增殖,提高细胞成骨分化水平,BMSCs复合PLGA膜可以促进牙周组织再生。
Objective To investigate the effect of bone marrow-derived mesenchymal stem cells(BMSCs)combined with PLGA membrane scaffold on periodontal tissue regeneration.Methods BMSCs of beagle dogs were isolated and cultured in vitro.The cell growth curve was detected by CCK8,and the purity of cells was detected by flow cytometry.PLGA membrane was prepared by solvent casting method,and the water absorption and dissolution rate of PLGA membrane were detected.The osteoblast differentiation rate was detected by alizarin red and ALP staining,and col1 and Runx2 were examined by WB.The periodontal tissue defect model of beagle dogs was prepared by operation.The periodontal tissue defect models were treated with saline injection,BMSCs injection,PLGA membrane transplantation and BMSCs combined with PLGA membrane transplantation,and the teeth without defect served as the control group.Results The proliferation level and osteogenic differentiation rate of BMSCs combined with PLGA membrane were significantly higher than those of monolayer culture.BMSCs combined with PLGA membrane could promote periodontal tissue regeneration in beagle dogs,and the effect was better than other groups.Conclusions PLGA membrane can provide a good microenvironment for BMSCs,promote cell proliferation and improve the level of osteogenic differentiation.BMSCs combined with PLGA membrane can promote periodontal tissue regeneration.
