摘要
目的探讨白介素-1(IL-1)信号通路在七氟烷预处理脊髓损伤(SCI)中的作用。方法按照是否进行SCI和是否进行预处理的原则将30只大鼠分为假手术组(S组)、脊髓损伤组(SCI组)和七氟烷预处理组(SEV组),每组10只。采用BBB评分评估三组神经功能缺损情况,运用HE染色计数存活神经元数量,通过免疫组化来标记小胶质细胞及IL-1β的数量,Western blot法测定IL-1β蛋白和NF-κB蛋白的表达水平。结果SCI组和SEV组的BBB评分低于S组,存活神经元数量少于S组(P<0.05);SEV组的BBB评分高于SCI组,存活神经元数量多于SCI组(P<0.05)。SCI组、SEV组的小胶质细胞和IL-1β免疫荧光表达数量多于S组(P<0.05);SEV组的小胶质细胞和IL-1β免疫荧光表达数量少于SCI组(P<0.05)。SCI组、SEV组的IL-1β和NF-κB蛋白表达量高于S组(P<0.05);SEV组的IL-1β和NF-κB蛋白表达量低于SCI组(P<0.05)。结论七氟烷预处理可能通过抑制IL-1信号通路减轻大鼠SCI。
Objective To investigate the role of interleukin-1(IL-1)signaling pathway in sevoflurane pretreatment for spinal cord injury(SCI).Methods Thirty rats were divided into sham operation group(S group),SCI group(SCI group)and sevoflurane pretreatment group(SEV group)according to the principle of whether spinal cord injury and pretreatment were given or not,with 10 rats in each group.BBB score was used to evaluate the neurological deficit of the three groups,HE staining was used to count the number of surviving neurons and immunohistochemistry was used to label the number of microglia and IL-1β.The expression levels of IL-1βprotein and NF-κB protein were determined by Western blot.Results BBB score in the SCI group and the SEV group was lower than that in the S group,and the number of surviving neurons was less than that in the S group(P<0.05);BBB score in the SEV group was higher than that in the SCI group,and the number of surviving neurons was more than that in SCI group(P<0.05).The immunofluorescence expressions of microglia and IL-1βin the SCI group and the SEV group were more than those in the S group(P<0.05);the immunofluorescence expressions of microglia and IL-1βin the SEV group were less than those in the SCI group(P<0.05).The protein expression levels of IL-1βand NF-κB in the SCI and SEV groups were higher than those in the S group(P<0.05);the protein expression levels of IL-1βand NF-κB in the SEV group were lower than those in the SCI group(P<0.05).Conclusion Sevoflurane pretreatment may alleviate SCI in rats by inhibiting IL-1 signaling pathway.
