摘要
目的建立同时测定黄芩茎、叶、花中野黄芩苷含量的高效液相色谱法。方法色谱柱为Agilent Zorbax SB-C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.4%磷酸水溶液(梯度洗脱),流速为1.0 m L/min,检测波长为335 nm,柱温为25℃,进样量为10μL。结果野黄芩苷质量浓度在24.38~243.80μg/m L范围内与峰面积线性关系良好(r=0.9999,n=6);精密度、稳定性、重复性试验结果的RSD均小于2.0%;黄芩叶中野黄芩苷的平均回收率为98.92%,RSD为0.90%(n=6)。结论该方法操作简便,专属性强,准确度高,重复性好,可用于黄芩茎、叶、花中野黄芩苷的含量测定。
Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of scutellarin in the stems,leaves and flowers of Scutellaria baicalensis.Methods The chromatographic column was Agilent Zorbax SB-C_(18) column(250 mm×4.6 mm,5μm),the mobile phase was methanol-0.4%phosphoric acid aqueous solution(gradient elution),the flow rate was 1.0 m L/min,the detection wavelength was 335 nm,the column temperature was 25℃,and the injection volume was 10μL.Results The linear range of scutellarin was 24.38-243.80μg/m L(r=0.9999,n=6).The RSDs of precision,stability and repeatability tests were less than 2.0%.The average recovery of scutellarin in the leaves of Scutellaria baicalensis was 98.92%with RSD of0.90%(n=6).Conclusion The method is simple,specific,accurate and reproducible,which can be used for the content determination of scutellarin in stems,leaves and flowers of Scutellaria baicalensis.
