出生后不同发育阶段小鼠耳蜗Connexin30的表达

摘要 目的研究缝隙连接蛋白30(Connexin30,Cx30)在出生后不同发育阶段小鼠耳蜗的表达分布,探讨其在听觉系统中的功能。方法选取出生后(postnatal day,P)1、5、7、10、14、21及30天即P1、P5、P7、P10、P14、P21和P30,共7个天龄段的小鼠,每组6只,通过免疫荧光染色和激光共聚焦显微镜检测Cx30在不同天龄小鼠耳蜗中的表达定位。结果P1组Cx30主要表达在大上皮嵴及耳蜗外侧壁;P5组Cx30广泛表达在血管纹基底细胞,螺旋缘出现Cx30的表达;P7组Corti器Deiters细胞开始表达Cx30;P10组Cx30表达在内凹细胞;P14组Cx30在大上皮嵴的表达消失,大部分螺旋韧带纤维细胞出现Cx30的表达;P21和P30组Cx30表达于Corti器支持细胞、血管纹基底细胞、螺旋韧带纤维细胞和螺旋缘。与P5组相比较,P7、P10、P14、P21和P30组耳蜗螺旋缘Cx30平均光密度值增高(P<0.05);与P7组相比,P10、P14、P21和P30 Deiters细胞Cx30平均光密度值明显增高(P<0.05);与P1组比较,P5、P7、P10、P14、P21和P30血管纹中Cx30平均光密度值均增高(P<0.05)。结论Cx30在不同鼠龄小鼠耳蜗表达和分布不同,其可能对小鼠听觉功能的建立和成熟具有十分重要的作用。 Objective To investigate the expression and spatial distribution of connexin30(Cx30)and its possible role in auditory system in the postnatal mouse cochlea.Methods The mouse cochleae were obtained at the 1,5,7,10,14、21 and 30 day after parturition.The expression of Cx30 in the mouse cochlea of 7 different age groups(n=6 each)was examined by immunofluorescence and laser scanning confocal microscope.Results At P1,Cx30 was mainly expressed in the greater epithelial ridge and cochlear lateral wall.At P5,Cx30 was widely distributed in the strial basal cells,and Cx30 began to be expressed in the spiral limbus.At P7,the expression of Cx30 occurred in the Deiters cells of the organ of Corti.At P10,CX30 expression was found in inner sulcus.At P14,Cx30 expression disappeared from the greater epithelial ridge,numerous fibrocytes located within the spiral ligament was immunopositive for Cx30.Between P21 and P30,Cx30 continued to be expressed in the supporting cells of the organ of Corti,basal cells of the stria vascularis,spiral ligament fibroblasts and spiral limbus.The average optical density of Cx30 in the spiral limbus at P5 was significantly different from those at P7,P10,P14,P21 and P30(P<0.05).The average optical density of Cx30 in the Deiters cells at P7 was significantly different from those at P10,P14,P21 and P30(P<0.05).The average optical density of Cx30 in the stria vascularis at P1 was significantly different from those at P5,P7,P10,P14,P21 and P30(P<0.05).Conclusion CX30 was dynamiclly expressed in the mouse cochlea before the onset of hearing,indicating that it might play a very important role in the development and maturation of the mouse cochlea.
