摘要
探究银耳多糖(Tremella Fuciformis Polysaccharide,TFP)对B16黑色素瘤细胞静脉注射所致肺转移模型的作用及其机理。将B16黑色素瘤细胞暴露于不同浓度(0、1.25、2.5 mg·mL^(-1))的TFP 24h,细胞重悬于PBS中,经ICR小鼠尾静脉注射,建立黑色素瘤模型。分别采用HE染色,Western Blot(WB)及免疫荧光染色探讨TFP对在体黑色素瘤细胞的作用机理。浓度为1.25、2.5 mg·mL^(-1)的TFP均能显著抑制B16黑色素瘤细胞肺转移的结节数目,并抑制过氧化物酶体增殖物激活受体(PPARγ)表达,增加CD86和CD206的表达。TFP对B16黑色素瘤细胞肺转移的抑制作用与抑制PPARγ表达从而招募巨噬细胞有关。
This study aims to investigate the effect and mechanism of Tremella fuciformis polysaccharide(TFP)on the B16 melanoma lung metastasis model.B16 melanoma cells were exposed to different concentrations of TFP(0,1.25,2.5 mg·mL^(-1))for 24 hours.The cells were resuspended in PBS and injected into the tail veins of ICR mice to establish the lung metastasis model.HE staining,WB and immunofluorescence were used to investigate the mechanism of TFP on melanoma.TFP significantly inhibited the lung metastasis of B16 melanoma cells,inhibited the expression of PPARγ,and increased the expression of CD86 and CD206.TFP inhibited the lung metastasis of B16 melanoma cells by inhibiting PPARγexpression and recruiting macrophages.
