摘要
Hippo信号通路可参与骨性关节炎的发生、发展,其效应因子Yes激酶相关蛋白/具有PDZ基序的辅转录激活子(YAP/TAZ)的下游基因Osterix可直接参与调控成骨细胞的转化,调控破骨细胞与成骨细胞的平衡。并且Hippo信号传导通路的效应蛋白YAP或TAZ可与多种引起骨关节炎的常见信号传导通路相交联,如转化生长因子超家族TGF-β/Smads、经典Wnt/β-Catenin信号转导通路和COX-Ⅱ,产生协同或抑制作用,进而参与骨性关节炎中的软骨退化,与骨赘形成和骨质破坏密切相关。此外,在骨性关节炎动物建模关节内注射Hippo信号通路相关干扰素,有效改善了骨性关节炎的发展,为临床上治疗骨性关节炎提供了新思路。
Hippo signaling pathway can participate in the development of osteoarthritis,and Osterix,the downstream gene of its effector protein YAP/TAZ,can directly participate in the regulation of osteoblast transformation and the balance between osteoclasts and osteoblasts.And the effector protein YAP or TAZ of the Hippo signaling pathway can be cross-linked with a variety of common signaling pathways leading to osteoarthritis,such as the transforming growth factor superfamily TGF-β/Smads,the classic Wnt/β-Catenin signaling pathway and COX-Ⅱ,which produces synergistic or inhibitory effects,is involved in cartilage degradation in osteoarthritis and is closely related to osteophyte formation and bone destruction.In addition,intra-articular injection of Hippo signaling pathway-related interferon in osteoarthritis animal modeling has effectively improved the development of osteoarthritis and provided new ideas for clinical treatment of osteoarthritis.
