响应面法优化铁皮石斛多糖的提取及其成分分析 被引量:14

摘要 采用响应面设计优化铁皮石斛多糖的提取纯化工艺,并结合苯酚-硫酸法对多糖含量进行测定。通过单因素试验,分析提取温度、提取时间、液料比和提取次数4个因素对铁皮石斛多糖得率及含量的影响。在此基础上,采用响应面设计优化铁皮石斛多糖的提取工艺,得最优工艺为:提取温度90℃,提取时间2.5 h,液料比60∶1,提取次数3次;在此条件下,铁皮石斛多糖含量的理论预测值为98.63%;对经优化法所得的铁皮石斛多糖成分进行理化分析,得其水分含量为7.39%,灰分含量为3.56%,无机盐含量为1.39%,氨基酸总量为0.67%。该方式所获得的铁皮石斛多糖含量高达85.33%,提取工艺简单便捷,得到的多糖成分稳定,富含多种人体必需氨基酸,为后续铁皮石斛多糖的功能活性研究奠定基础。 The extraction and purification process of polysaccharides from Dendrobium officinale was optimized by using the response surface method(RSM),and the content of polysaccharides was determined by using phenol-sulfuric acid method.The effects of extraction temperature,extraction time,liquid-solid ratio and extraction frequency on the yield and content of polysaccharides from D.officinale were analyzed by single factor experiment.Response surface design was then used to find the optimal extraction process of polysaccharides from D.officinale.The theoretical prediction value of polysaccharides content extracted from D.officinale was 98.63%under the optimum extraction conditions:extraction temperature 90℃,extraction time duration 2.5 h,liquid-solid ratio 60:1,and extraction frequencies 3.Physicochemical analysis of the components of the polysaccharides extracted from D.officinale showed that the polysaccharides contained water 7.39%,ash 3.56%,inorganic salt 1.39%,and amino acids 0.67%.The polysaccharides content extracted from D.officinale under the optimum conditions was as high as 85.33%,and the extraction process was easy and simple.The extracted polysaccharides were stable in composition and rich in essential amino acids,which facilitates further research in their functional activities.
