摘要
目的:对比研究长梗绞股蓝总皂苷和绞股蓝总皂苷的调血脂作用。方法:大鼠90只,随机分组,分别为正常对照组、模型组、阳性对照药组(1 mg·kg^(-1))、绞股蓝总皂苷高、中、低剂量组(60,30,15 mg·kg^(-1))、长梗绞股蓝总皂苷高、中、低剂量组(60,30,15 mg·kg^(-1))。大鼠高脂饲料饲养造模后灌胃(ig)给予相应药物,连续6周。测定大鼠体重、肝脏指数、血脂水平、谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate transaminase,AST)含量,HE染色检测肝脏脂肪变性情况。结果:与正常对照组相比,模型组大鼠的体重和肝脏指数、血清总胆固醇(total cholesterol,TC)、三酰甘油(triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、ALT和AST含量增加,高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)降低(P<0.05或P<0.01),肝脏可见明显脂肪变性。与模型组相比,绞股蓝总皂苷可降低血清TC和ALT含量,长梗绞股蓝总皂苷可显著降低TC,TG,AST,ALT含量,提高HDL-C含量。长梗绞股蓝和绞股蓝总皂苷高剂量组均可减轻大鼠肝脏脂肪变性情况。经相关性分析发现,血清TC,TG,AST与总皂苷来源相关。结论:长梗绞股蓝总皂苷和绞股蓝总皂苷均显示出降脂和肝脏保护的作用,但长梗绞股蓝总皂苷降血脂和肝脏保护作用更显著。
Objective:To compare the lipid-regulating effects of long gypenosides and gypenosides.Methods:Ninety male rats were randomly divided into control group,model group,positive group(1 mg·kg^(-1)),gypenosides groups(60,30 and 15 mg·kg^(-1))and long gypenosides groups(60,30 and 15 mg·kg^(-1)).The hyperlipidemia model was established by feeding high-fat diets,then all rats were intervened with corresponding medicines for 6 weeks.The body weight,liver index,levels of blood lipid,alanine aminotransferase(ALT)and aspartate aminotransferase(AST)were determined.HE staining was conducted to evaluate the histopathological changes of livers.Results:Compared with the control group,the body weight,liver index,total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),ALT and AST in the model group were significantly increased.The high-density lipoprotein cholesterol(HDL-C)level was significantly decreased.Fatty degeneration was obviously observed in liver tissue.Compared with the model group,gypenosides significantly decreased TC and ALT levels in serum.Long gypenosides significantly reduced the levels of TC,TG,AST and ALT,increased the level of HDL-C.High dose long gypenosides and gypenosides relieved the fatty degeneration.The results of correlation analysis showed that the levels of TC,TG and AST in serum correlated with plant origin.Conclusion:Long gypenosides and gypenosides can reduce blood lipid and have positive effect on liver tissue of hyperlipidemia rats,especially long gypenosides.
